Show moreAtlantic spotted dolphins (Stenella frontalis) and bottlenose dolphins (Tursiops truncatus) are two sympatric species resident to the Bahamas. The visibility of the Bahamian water provided a unique opportunity to study spotted dolphin communication during aggression. This study’s main focus was to decipher any similarities or differences in the behaviors used by spotted dolphins during interspecific and intraspecific aggression. Both similarities and differences were discovered. Biting, following, and chasing behavioral events were used more during interspecific aggression, while the display behavioral class was used more than the contact behavioral class during intrabut not interspecific aggression. This study showed that spotted dolphins use more energy intensive and risky behaviors when fighting interspecifically. This could result from having to fight and defend females from a larger species, trying to avoid sexual harassment from bottlenose males, or needing to use behaviors that are more overt and easily understood during interspecies communication.

A wide range of studies have focused on Msr both in vivo and in vitro using a variety of model organisms. More specifically, studies have noted numerous processes affected by the overexpression, under expression, and silencing of MsrA and MsrB. Collectively, the results of these studies have shown that Msr is involved in lifespan and the management of oxidative stress. More recent evidence is emerging that supports existing biological functions of Msr and theorizes the involvement of Msr in numerous biological pathways.

In this study, unhatched eggs were collected from the Boca Raton, Florida beach and developmental stage at embryonic death determined. Elevated nest temperatures increased embryonic mortality, and the most significant relationship was between mortality and the percent of time embryos were exposed to temperatures above 34°C. Loggerhead turtles exhibited higher rates of mortality compared to green turtles at temperatures above 34°C. Only loggerhead nests were exposed to inundation, but embryonic mortality did not differ from noninundated nests. Beach profiles across the nesting season were also determined. A major storm altered the beach more in areas of coastal development; however, this was impacted by a nourishment project and the presence of a structured inlet. Future management strategies may need to protect sea turtle nests from extended periods at elevated temperatures.

In this dissertation, I studied territoriality, behavioral plasticity and syndromes, song learning, and natal dispersal in the Bachman’s sparrow (Peucaea aestivalis), an imperiled songbird that is understudied in many aspects of its behavior. In chapter one, I provide a background of behavioral ecology, the topics covered in this dissertation, and the Bachman’s sparrow. In chapter two, I determine if the ideal despotic distribution applies to Bachman’s sparrows. In chapter three, I examine whether two common personality traits, aggressiveness and boldness, are consistent over time and form a behavioral syndrome, or if they change in relation to environmental variables. My last chapter infers song learning and natal dispersal strategies in Bachman’s sparrows from examining patterns song-type sharing over geographic distance.

Show moreIn the round worm C. elegans, it has recently been shown that autophagy, a highly conserved lysosomal degradation pathway that is present in all eukaryotic cells, is required for maintaining healthspan and for increasing the adult lifespan of worms fed under dietary restriction conditions or with reduced IGF signaling. It is currently unknown how extracellular signals regulate autophagy activity within different tissues during these processes and whether autophagy functions cell-autonomously or nonautonomously. We have data that for the first time shows autophagy activity in the neurons and intestinal cells plays a major role in regulating adult lifespan and the longevity conferred by altered IGF signaling and dietary restriction, suggesting autophagy can control these phenotypes cell non-autonomously. We hypothesize that autophagy in the neurons and intestinal cells is an essential cellular process regulated by different signaling pathways to control wild type adult lifespan, IGF mediated longevity and dietary restriction induced longevity. Excitingly we also have found that in animals with reduced IGF signaling autophagy can control longevity in only a small subset of neurons alone. Autophagy in either specific individual chemosensory neurons or a small group of them is completely sufficient to control IGF mediated longevity. This work provides novel insight to the function and regulation of autophagy which will help shed light on understanding this essential process in higher organisms, including mammals.

The objectives of my study were to expand knowledge on the behavioral ecology of lesula and evaluate lesula’s sensitivity to hunting threats. The main findings from my study included: confirmation of terrestriality and diurnality, births clustering during the wet season, social group living of one male and multiple females, and high impact of hunting on group size. By studying the latest added species to the Cercopithecini tribe, my thesis helps to better understand the ecological diversity occurring within this radiation of African primates and contributes to the species’ long-term conservation efforts.

Show moreThis study investigated a resident community of Atlantic spotted dolphins (Stenella frontalis) on Little Bahama Bank (LBB) in the Bahamas utilizing a noninvasive molecular approach. Genetic template material was collected and extracted from fecal material of S. frontalis. Fine-scale population structure was found within LBB according to mitochondrial DNA (mtDNA) and microsatellites (Fst = 0.25317, P < 0.0001 and Fst = 0.04491, P < 0.0001, respectively). Three main social clusters (North, Central, South/Roam) exist on LBB and all clusters were found to be genetically distinct according to microsatellite analyses. Mitochondrial haplotypes revealed North and South/Roam were not differentiated, but Central was different from both. When separated by sex, males were less genetically structured than females. Males showed no evidence of structure according to Ost or Rst., Females of all clusters were differentiated according to microsatellites whereas mtDNA revealed the same pattern in females as was seen for the total population. The structuring patterns of the sexes clearly indicate a pattern of male dispersal and female philopatry for the LBB population. Genetic investigation of mating revealed patterns in the mating system of S. frontalis on LBB. Genotypes of females and offspring were analyzed and revealed that more than two males were required to explain the progeny arrays, indicating promiscuous mating among females. In addition, paternity assessment assigned seven males as fathers to ten of 29 mother-calf pairs. A pattern of reproductive skew according to age was revealed because reproductively successful males were in the oldest age class at the estimated time of conception of the calves., Patterns in social cluster mating revealed that males from the Central cluster sired offspring with females from both the Central and North clusters, while Roaming males sired offspring with South and Central females indicating that males mate within their social cluster or with females from the next closest cluster. The study has important implications for cetacean research, specifically delphinids. Fine-scale population structure and mating patterns of male and female S. frontalis were revealed through noninvasive methodology presenting a valuable genetic framework with which to support ongoing investigations of life history, behavior, communication and social structure.

Show moreUnder the expected warmer temperatures due to climate change, sea turtle embryos may be subjected to thermal conditions detrimental to nest success and hatchling quality; one trait which may be negatively affected is cognitive ability. In this study, loggerhead sea turtle eggs were acquired from Boca Raton, FL and lab incubated under two female-producing temperatures: an “optimal” temperature of 31°C and a sublethal temperature of 33°C. Cognitive ability of post-hatchlings, assessed via associative learning and reversal was investigated using a y-maze. The sublethal temperature decreased incubation duration, hatch success, hatchling growth rates and produced smaller hatchlings with significantly more scute anomalies. Hot hatchlings performed worse on the reversal, taking longer to train, and thus hint at an effect of incubation temperature on cognitive flexibility in loggerhead turtles. With temperatures rising on beaches in South Florida, this study provides evidence of further potential threats to hatchling quality and potentially even survival.

Show moreNeurons are able to maintain membrane potential and synaptic integrity by an intricate equilibrium of membrane transporter proteins and ion channels. Two membrane proteins of particular importance in the vertebrate retina are the excitatory amino acid transporters (EAATs) which are responsible for the reuptake of glutamate into both glial and neuronal cells and the sodium potassium chloride cotransporters (NKCCs) that are responsible for the uptake of chloride ions into the cell. NKCCs are electro-neutral with the uptake of 2 Cl- coupled to an exchange of a potassium and Na+ ion into the cells. Therefore, there is little change of cell membrane potential in the action of NKCCs. In this study the localization and function of EAATs in the distal retina is investigated. Whole cell patch clamp recordings in lower vertebrate retina have demonstrated that EAAT2 is the main synaptic EAATs in rod photoreceptors and it is localized to the axon terminals. Furthermore, the action of the transporter seems to be modified by intracellular calcium concentration. There is also evidence that EAAT2 might be regulated by feedback from the neuron network by glycinergic and GABAergic mechanisms. The second half of this study investigates expression of NKCCs in the retina by western blot analysis and quantitative polymerase chain reaction. There are two forms of NKCCs, NKCC1 and NKCC2. NKCC1 is mostly expressed in the central nervous system and NKCC2 was thought to only be expressed in the kidneys. NKCC1 is responsible for the majority of chloride uptake into neuronal and epithelial cells and NKCC1 is expressed in the distal retina where photoreceptors synapse on second order horizontal and bipolar cells. This study found the expression of NKCC1 in the distal retina to be regulated by temporal light and dark adaptation. Light adaptation increased phosphorylated NKCC1 expression (the active form of the cotransporter). The increase in NKCC1 expression during light adaptation was modulated by dopamine. Specifically, a D1 receptor agonist increased phosphorylated NKCC1 expression. Dopamine is an essential chemical and receptor known for initiating light adaptation in retina. Finally, an NKCC1 knockout mouse model was examined and it revealed that both forms of NKCC are expressed in the vertebrate retina.

Show moreAnthropogenic contaminants in the marine environment often biodegrade slowly, bioaccumulate in organisms, and can cause reproductive, immune, and developmental effects on wildlife; however, their effects on cetacean health and behavior have not been extensively studied. This study aims to expand knowledge concerning concentrations and biological effects of endocrine disrupting contaminants and essential and non-essential inorganic elements in stranded cetaceans. We evaluated tissue samples and pathology data from 66 odontocetes that stranded in the southeastern United States during 2012– 2017. Using mass spectrometry blubber samples were analyzed for five endocrine disrupting contaminants (atrazine, bisphenol-A, diethyl phthalate, nonylphenol ethoxylate, triclosan), and liver samples were analyzed for 12 inorganic elements (As, Cd, Co, Cu, Mg, Mn, Fe, Pb, Hg, Se, Tl, Zn). Results from this study demonstrate that exposure to certain contaminants may be associated with subtle or sublethal cellular changes in free-ranging marine mammals that could contribute to health declines or stranding.

Show moreThe adult rodent olfactory system provides an excellent model for the study of the developing nervous system due to its constant integration of new neurons and establishment of new synapses throughout life. This allows for adaptive structural responses to sensory experience that continuously shape functional circuits to fit the behavioral needs of the animal. The cellular mechanisms underlying the anatomical plasticity of the adult olfactory system are not well understood, but neurotrophins, long recognized as crucial to the early development of neuronal circuits, are attractive candidate molecules in this capacity. Brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF), two members of the neurotrophin family, have received attention in terms of their anatomical distribution and physiological functions in the olfactory system.

Topotecan Hydrochloride (Hycamtin) is an FDA-approved chemotherapy drug, primarily used for secondary treatment of ovarian,cervical and small cell lung cancers. This study was to demonstrate the potential anticancer activities and synergy of topotecan-genistein combination in LNCaP prostate cancer cells. The potential efficacy and mechanism of topotecan/genistein-induced cell death was investigated... Results: The overall data indicated that i) both genistein and topotecan induce cellular death in LNCaP cells, ii) topotecan-genistein combination was significantly more efficacious in reducing LNCaP cell viabiligy compared to either genistein or topotecan alone, iii) in all cases, cell death was primarily through apoptosis, via the activation of the intrinsic pathway, iv) ROS levels were increased and VEGF expression was diminished significantly with the topotecan-genistein combination treatment, v) genetic analysis of topotecan-genistein treatment groups showed changes in genetic expression levels in pathway specific apoptotic genes.... Conclusion: Treatments involving topotecan-genistein combination may prove to be an attractive alternative phytotherapy of adjuvant therapy for prostate cancer.

Cells were exposed to single and combined treatments with the phytochemiclas for 48 and 72 hours. Cell viability was assessed using the MTT bioassay. Apoptosis induction was assessed using acridine orange ethidium bromide and rhodamine 123 ethidium bromide fluorescence assays. Both singe and combination treatments induced dose- and time-dependent apoptotic cell death in MCF-7 cells. The percentage of apoptosis was higher in combination treatments than single treatments with either phytochemical, although the difference was not statistically significant. The combination of genistein and anthocyanin extract peaked in efficacy at 48 hours of treatment, to exhibit significantly greater (P<. O5) dose- and time-dependent cell cytotoxicity than single treatments. This study reveals potential chemopreventive implications for the complementary effects of genistein and anthocyanin extract.
