Show moreThe Computer Tomography (CT) scanned images are very important for the Treatment Planning System (TPS) to provide the electron density of the different types of tissues that the radiation penetrates in the path to the tumor to be treated. This electron density is converted to an attenuation coefficient, which varies with tissue for each structure and even varies by the tissue volume. The purpose of this research is to evaluate the CT numbers, and convert them into relative electron densities. Twenty-five patients’ data and CT numbers were evaluated in the CT scanner and in Eclipse and were converted into relative electron density and compared with each other. The differences between the relative electron density in the Eclipse was found to be from 0 up to 6% between tissue equivalent materials, the final result for all equivalent tissue materials was about 2%. For the patients’ data, the percentage difference of CT number versus electron density was found to be high for high relative electron density organs, namely the final average result for the spine was 8%, less for pelvis, and less for rib while for the other organs it was even less. The very lowest was 0.3% compared with 1% which is acceptable for clinical standards.

Show moreThe purpose of this research is to explore and investigate the biophysical properties of living cells using microfluidics based electrical impedance sensing (EIS) technique. It provides a non-invasive approach to detect label-free biological markers in the regulation of cellular activities even at a molecular level. We specifically focus on the development, testing, and theoretical modeling of electrical impedance spectroscopy for neuroblastoma cells and endothelial cells. First, we demonstrate that the EIS technique can be used to monitor the progressive mitochondrial fission/fusion modification in genetically modified human neuroblastoma cell lines. Our results characterize quantitatively the abnormal mitochondrial dynamics through the variations in cytoplasm conductivity. Secondly, we employ a real time EIS method to determine the biophysical properties of the junctions which join one endothelial cell with one another in a monolayer of endothelial cells. In particular, we examine the role of the protein, c-MYC oncogene, in the barrier function. Our results show that the downregulation of c-MYC oncogene enhances the endothelial barrier dysfunction associated with inflammation. Finally, we measure and find that the electrical admittance (the reciprocal of the impedance) of the monolayer of endothelial cellular networks exhibits an anomalous power law of the form, Y ∝ ωα, over a wide range of frequency, with the value of the exponent, α, depending on the severity of the inflammation. We attribute the power law to the changes of the intercellular electric permeability between neighboring endothelial cells. Thus, the inflammation gives rise to relatively smaller values of α compared to that of the no-inflammation group. Furthermore, we propose a simple percolation model of a large R-C network to confirm the emergent of power law scaling behavior of the complex admittance, suggesting that the endothelial network behaves as a complex microstructural network and its electrical properties may be simulated by a large R-C network.

For the large electron fields sizes, we had to use the 2D water tank. All the collected data was converted into a file type accepted by the planning system (Eclipse) and subsequently imported there. Treatment plans were generated using multiple forms of planning to verify the viability and quality of the beam data commissioned.

We analyze the mean-field treatment of the model for electrolyte of arbitrary valences, and then beyond, where correlation effects are systematically taken into account in a loop expansion. In particular, we compute exactly various one-loop quantities, including electrostatic potentials, ion distributions, and chemical potentials, not only for symmetric (1, 1) electrolyte but also for asymmetric (2, 1) electrolyte, and make use of these quantities to address charge regulation at the one-loop level. We find that correlation effects give rise to various phase transitions in the adsorption of ions, and present phase diagrams for (1, 1) and (2, 1) electrolytes, whose distinct behaviors suggest that charge regulation, at the one-loop level, is no longer universal but depends crucially on the valency of the ions.

When used as carriers of quantum information, they have no equal. For the processing of quantum information, single photons have proven difficult to scale beyond the order of ⇠ 10 photons. The lack of single-photon-level interaction has led to creative approaches which rely on postselection to filter the possible measured outcomes to those which appear as though interaction occurred. This approach of post-selection leans heavily on the ability to not only generate but also detect scores of single photons simultaneously and with near perfect efficiency. Our work relaxes the emphasis which has been placed on single photons for quantum information processing to that of states with, in principle, an arbitrary number of photons. Central moment expectations on two-mode squeezed states are shown to exhibit post-selection behavior which reflects the single-photon counterpart. These measures are proven to be robust to loss and return entangled state statistics on average. With naive estimation of the central moment, states with ~ 20 modes are within reach with current technology, closing the gap between quantum states which can and cannot be classically simulated.

Creating an improved BAO can decrease the amount of time a dosimetrist spends on making a treatment plan, improve the treatment quality and enhance the tools an inexperienced dosimetrist can use to develop planning techniques. Using empirical data created by experienced dosimetrists from 69 patients treated for lung cancer, the most frequently used gantry angles were applied to four different regions in each lung to gather an optimal set of fields that could be used to treat future lung cancer patients. This method, given the moniker FAU BAO, is compared in 7 plans created with the Eclipse BAO choosing 5 fields and 9 fields. The results show that the conformality index improved by 30% or 3% when using the 5 and 9 fields. The conformation number was better by 12% from the 5 fields and 9% from the 9 fields. The organs at risk (OAR) were overall more protected to produce fewer nonstochastic effects from the radiation treatment with the FAU BAO.

Show more1In this dissertation we work out in detail a new proposal to define rigorously a sector of loop quantum gravity at the diffeomorphism invariant level corresponding to homogeneous and isotropic cosmologies, and propose how to compare in detail the physics of this sector with that of loop quantum cosmology. The key technical steps we have completed are (a) to formulate conditions for homogeneity and isotropy in a diffeomorphism covariant way on the classical phase space of general relativity, and (b) to translate these conditions consistently using well-understood techniques to loop quantum gravity. To impose the symmetry at the quantum level, on both the connection and its conjugate momentum, the method used necessarily has similiarities to the Gupta-Bleuler method of quantizing the electromagnetic field. Lastly, a strategy for embedding states of loop quantum cosmology into this new homogeneous isotropic sector, and using this embedding to compare the physics, is presented.

It includes five modules to perform different QA tests: Star Shot analysis, Picket Fence test, Winston-Lutz test, MLC log file analysis, and verification of light & radiation field coincidence. We also pay attention to quality assurance of 6DOF treatment couch that plays a very important role in radiation therapy. We developed an Arduino based 3D printed 6DOF robotic phantom to check the accuracy of the treatment couch. This robotic phantom was experimentally validated under clinical standards, and customizable upon requirements of the quality assurance Task. The current features of this robotic phantom open development opportunities beyond the specific couch application, such as organs motion simulation.

Associated tolerance levels were established based on these sensitivities and guidelines published in recent American Association of Physics in Medicine (AAPM) task group reports. The output has remained within the 3% tolerance and the range was within ±1mm. Spot position has remained within ±2mm. This daily QA procedure is quick and efficient with the time required for setup and delivery at less than 10 minutes.

However, a small error in dose calculation may lead towards undesired greater uncertainties in planed doses. This project aims to create a simulation model of Varian ProBeam Compact using the GEANT4 Monte Carlo simulation tool kit. Experimental data from the first clinical ProBeam Compact system at South Florida Proton Therapy Institute was used to validate the simulation model. A comparison was made between the experimental and simulated Integrated Depth-Dose curves using a 2%/2mm gamma index test with 100% of points passing. The beam spot standard deviation sizes (s!) were compared using percent deviation. All simulated s! matched the experimental s! within 2.5%, except 70 and 80 MeV. The model can be used to develop a more comprehensive model as an independent dose verification tool and further investigate dose distribution.

Show moreA detailed dosimetric comparison between Inverse Planning by Simulated Annealing (IPSA) and Dose Points (DP) optimized treatment plans has been performed for High Dose Rate (HDR) brachytherapy of skin lesions using Freiburg Flap applicator in order to find out whether or not IPSA offers better clinical dosimetric outcomes for lesions categorized into four different curvatures. Without compromising target coverage, IPSA reduced the volume of Planning Target Volume (lesion) that received at least 125% of the prescription dose on average by 41%. It also reduced the volume of the healthy skin surrounding the lesion that receives at least 100% of the prescription dose on average by 42%. IPSA did not show any advantage over DP in sparing normal structures underlying the lesions treated. Although DP optimization algorithm has been regularly used at Lynn Cancer Institute for HDR brachytherapy of skin lesions, recent upgrades in IPSA software have made IPSA more amenable to rapid treatment planning and therefore IPSA can be used either in place of DP or as its alternative.

As a result, the actual dose delivered may vary from the original treatment plan. The purpose of this study was (a) to evaluate the dosimetric consequences of the parotid glands during the course of treatment, and (b) to determine if there would be an optimal timeframe for replanning. Nineteen locally advanced HN cancer patients underwent definitive IMRT. Each patient received an initial computerized tomography simulation (CT-SIM) scan and weekly cone beam computerized tomography (CBCT) scans. A Deformable Image Registration (DIR) was performed between the CT-SIM and CBCT of the parotid glands and Planning Target Volumes (PTVs) using the Eclipse treatment planning system (TPS) and the Velocity deformation software. A recalculation of the dose was performed on the weekly CBCTs using the original monitor units. The parameters for evaluation of our method were: the changes in volume of the PTVs and parotid glands, the dose coverage of the PTVs, the lateral displacement in the Center of Mass (COM), the mean dose, and Normal Tissue Complication Probability (NTCP) of the parotid glands. The studies showed a reduction of the volume in the PTVs and parotids, a medial displacement in COM, and alterations of the mean dose to the parotid glands as compared to the initial plans. Differences were observed for the dose volume coverage of the PTVs and NTCP of the parotid gland values between the initial plan and our proposed method utilizing deformable registration-based dose calculations.
