Earth’s environment has been undergoing unprecedented rates of degradation during the Anthropocene paradigm. Current projections for the near future show climate change producing grim outcomes for most habitable parts of the world. Show moreEarth’s environment has been undergoing unprecedented rates of degradation during the Anthropocene paradigm. Current projections for the near future show climate change producing grim outcomes for most habitable parts of the world. This thesis defends the argument that in order to adequately address the state of the environment, humanity must experience a shift in collective consciousness away from the current philosophical paradigm, and instead adopt a paradigm that enables a common mindset regarding the place of humans within the natural environment. Various forms of spiritual ecology are explored: deep ecology, biblical eco-theology, and eco-feminist theology. These positions are explored in order to introduce a framework necessary to achieve the collective shift in consciousness required to address environmental issues: a Spiritual Framework of Organic Oneness, which includes components of spiritual ecology and earth-centered religious traditions.

Our rapid method – “In-the-zone” (ITZ) employs a) the size of zones of inhibition b) identity of bacteria in the zone c) number of resistant clones within zones of inhibition and d) type of antibiotic to compute ARI. Results can be obtained within 24 - 48 hrs with very little effort. This semester, we repeated the test and validated the weighting of various parameters. The ITZ assay yield consistent and reproducible data on zones of inhibition of bacteria community. The diameter of inhibition zones was the most important parameter distinguishing resistomes of varying ARI. Landfill, manured garden, and waste plant samples scored highest in antibiotic resistance index compared to beach sand.

Show moreGenomics has been revolutionized by improved sequencing technologies, allowing for the detailed exploration of microbial dark matter and complex microscopic ecosystems. The bottleneck in genomic workflows has shifted from high-throughput sequencing to data analysis. This dissertation developed the Florida Center for Coastal and Human Health Shotgun Metagenomics Workflow (FCHsm) that is easy to use and tailor to unique datasets. This work acts as the beta-testing for the workflow, as it analyzes disparate biomes (environmental and host microbiomes) at varying sequencing depths (shallow and deep). FCHsm was used to resolve molecular dynamics and mine trans-kingdom metagenomes for secondary metabolic biosynthetic gene clusters (BGCs) in two marine environments— Indian River Lagoon toxic harmful algal blooms (IRL HABs) and the medicinal Leiodermatium sponge holobiont. First, an in silico mock dataset was analyzed to benchmark the FCHsm workflow. Sourmash, coupled with the Genome Taxonomy Database, outcompeted the other taxonomic profilers by accurately predicting the size of the mock metagenome (450 genomes) and recalling the highest number of species (82 %) and strains (44 %). Nonpareil calculated the sequencing effort needed for 100 % coverage for all the datasets and correctly estimated the 75 Gbp of sequencing needed for almost 100 % coverage of the mock metagenomes (99.5 %). Next, the trans-kingdom metagenomes of the IRL were explored, and potential HAB biomarkers were identified.

For example, Huxley’s Brave New World and Brave New World Revisited illustrate with contempt a society that had renounced personal individuality and rejected freedom, choosing a drugged totalitarian state set adrift from any sense of morality. Huxley’s work is a familiar touchstone, in that it presupposed a world that seems increasingly real to us, even though a faithful portrayal of the future was hardly Huxley’s intent. Bradbury’s Fahrenheit 451 soberly reminded the world of the wages of intolerance to divergent ideas, in envisioning a government solidifying its hold on power by banishing the spectrum of human emotion in literature and mass media. Forster’s ambitious short story “The Machine Stops” countered the naiveté of a positive equal utopia, with a world cruel in its homogenization and dependent on a deified machine for all facets of its existence. In each instance, government is an ominous, questionable character. Technology, in these texts and in today’s world, is a foundational element with muddled aims—a rich virtual society on one hand, but frightening levels of assimilation, control, and loss of interpersonal communication and privacy in the crumbling arena of the real on the other. In this article, a future technology-led existence is examined through the lenses of these fictional works.
