Show moreGold is a highly prized metal whose ore deposits are continually being mined and depleted. With the increasing scarcity of continental gold ores, gold miners and scientist are turning to the sea as one alternate source of this precious metal. But the problem arises in determining which parts of the ocean to mine. Ideally the area with the greatest concentration of gold would be the best place to start but determining an accurate concentration of gold in a sample of seawater is difficult for various reasons. The main reason being gold's extremely low concentration in seawater, averaging at about13 [sic] parts per trillion. This study deals with detecting trace amounts of gold in seawater. The first step in the analysis involves pre-concentrating gold in seawater using an ion exchange resin, Chelexª -100, and optimizing the experimental conditions. Then the chromatographic fractions are analyzed using a graphite furnace atomic absorption spectrometer. The results from several trials are presented in this study.

Show moreDynactin is a multi-subunit protein complex that is the required cofactor for the cytoplasmic motor protein, dynein. Dynactin is critical for cytoplasmic activities such as vesicle transport, cytoskeletal organization, membrane organization and mitotic progression as it serves to increase processivity of dynein. The main subunit in focus for this study is the sidearm extension p150Glued. On the exposed end (N-terminus) lie two different microtubule binding motifs: CAP-Gly and Basic. CAP-Gly has a high binding affinity for microtubules, while Basic binds the microtubules quite weakly. It is at this location that dynactin is proposed to assist in the processivity of transporting cargo long distances in the cell, by providing additional support for the dynein molecule as it progresses along the microtubule. Analysis of the effects of overexpression of these motifs, alone or in tandem, suggested that there was no disruption of the dynein/dynactin interaction, but its ability to act in a processive manner had been perturbed. Effects on non-dynamic processes, such as Golgi localization and microtubule organization were generally weak, but effects were greater on y-tubulin organization. While this could suggest a defect in centrosome organization, we propose that it instead shows that cargo motility in the affected cells is defective, and that transport of centrosomal proteins to the centrosome is interrupted, suggesting a broader cargo-transport defect may be evident in these cells.

Reef health worldwide is declining due to many factors: e.g. pollution, sedimentation, dynamite fishing, and global warming. Working with the Planetary Coral Reef Foundation and the crew aboard the R/V Heraclitus, I examined coral health and fish diversity in the Solomon Islands, Melanesia. In 2006, I surveyed coral and fish on Sagharughombe reef, Solomon Islands using SCUBA and compared the results with surveys done in 2000 and 2002. Coral surveys indicated a significant decline in health over the six-year period. Fish surveys found no clear trend in species abundances and species richness, but a significant decline in species diversity over the years. The decline in coral health we observed may have contributed to diminished fish diversity. To protect marine biodiversity, coral reefs must be better protected and declines in coral health must be halted.

Western anti-FGM discourse is predicated upon the belief that the body must be kept in its "natural" or "pristine" state, and that any practice which violates the body's natural "perfection" is mutilation. Show moreIn this thesis, I argue that all bodies are material-semiotic entities, produced by both natural and cultural processes. Western anti-FGM discourse is predicated upon the belief that the body must be kept in its "natural" or "pristine" state, and that any practice which violates the body's natural "perfection" is mutilation. Implied by this discourse is the false notion that Western bodies are given and left unaltered. By drawing comparisons between Western genital practices and non-Western genital practices, I undermine the ideology that erases the working of culture on Western bodies while highlighting the "mutilating" powers exercised on the bodies of Others. Current imperialist hegemony perpetuates the view of African women as passive victims of barbaric tradition in need of rescuing by Western liberated women. We must, instead, work toward theories that account for differences in experience and history, rather than those which posit universal understandings of patriarchy and domination.

This study attempted to induce an effect opposite to anxiety interpersonal connectedness-using a video clip. This feeling of connectedness was anticipated to decrease the need for the defenses described by terror management theory, and more specifically to lower the need for cultural worldview defense. Participants were assigned to one of two video conditions: the control video condition (K-Web) or the experimental video condition (42-Up). After the video clip was shown, participants judged a list of social transgressions and rated the amount of punishment they felt the transgressor deserved. Individuals with high levels of self-esteem and positive affect were found to be more punitive than their counterparts, but the amount of punishment doled out to the transgressors was lower in the experimental video groups than in the control video groups.

R. mangle is a completely viviparous species, and as such, maternal conditions have a strong effect on propagules. My study compares the initial establishment and growth of R. mangle propagules collected from and planted in both impounded and non-impounded sites.

Currently, a cocktail of drugs administered via injection (HAART) has some efficacy in treating HIV, but does not eradicate HIV from infected individuals and has long-term side effects. In addition, drug-resistant variants of HIV are emerging. In an effort to help develop orally administered anti-HIV drugs, we examined membrane permeability of four scaffold peptides (synthesized by a researcher at Scripps Florida) into T-cells. One peptide (KE1-72A) entered cells with 100% efficiency; a second (KE1-72B) showed minimal cell penetration. Two other peptides (KE1-72C and KE1-72D), when chemically conjugated to an HIV fusion inhibitor, also showed minimal cell penetration. Further research is needed to determine whether the peptide KE1-72A may potentially be useful in orally delivered anti-HIV drugs.

Show moreThe purpose of this paper is to break through the complexity of the NAFTA negotiations in order to reveal some of the contentious issues from three stages of the NAFTA bargaining process: the fast track agreement, the negotiations under President Bush, Sr., and the side payments under President Clinton. Putnam's two-level game theory will help describe how the interests of business, environmental groups, and labor unions influenced the outcome of the NAFTA through their respective win-sets, domestic and international power relations, and side agreements. Extrapolating from Putnam's model and the success in NAFTA bargaining, we can predict that the ongoing Free Trade Area of the Americas (FTAA) negotiations are more likely to succeed if international representatives strategize to create favorable conditions for domestic ratification through understanding the domestic constituencies and win-sets of the players.

Scholars who lament the decline of the western canon claim that a massive wave of new untested works - largely written by women and people of color - have replaced the works of dead white men leaving our students ill-prepared. While most of the scholarship in this area has been written in the area of English literature departments, this project focuses on the field of Hispanic Literature. If identity politics has challenged the canon in the university, it is expected that within identity-based disciplines the infiltration of politics should be substantial. To test the politicization of the university, I examined 38 Hispanic Literature survey courses from a variety of American universities. I found a high degree of consistency among these syllabi and concluded that critics of the university have at best overstated their case.

Members of Japanese society perpetuate their collective memory through the utilization of Japanese media to include past and present Japanese name alterations. I explain the reasons behind name alterations, and how knowledge and use of the naming rituals continue through the collective agency of contemporary Japanese media such as anime and manga. Each anime and manga example correlates to past and present naming rituals. Social name alterations occur at birth, genpuku, marriage, and changes in levels of skill. Political alterations occur from hostage exchange or adoption, change in ideologies, occupational change, or the assumption of new roles from a higher-ranking member of society. While members of Japanese society learn naming traditions from daily interactions with other people, media such as anime and manga reinforce expected behavior and customs.

Show moreTalin is a ubiquitous, high-molecular-weight, flexible protein that plays a critical role in focal adhesions by activating, as well as connecting, integrins to the actin cytoskeleton. Talin's inactive auto-inhibitory state is speculated to be one of its modes of regulation inside the cell and is achieved through its head-tail interactions. In order to decipher the stability of this interaction, the head domain (residues 206-405) was cloned into a modified pET28m vector while the tail domains (residues 1654-2344 and 2225-2344) were cloned into the pET32a vector to obtain octa-histidine tagged and un-tagged peptide, respectively. Neither co-expression nor pull-down using the His-tagged head domain was successful in purifying a stable head-tail complex. Our results indicate rather weak interactions between the talin head and rod domains and hence, under our experimental conditions, do not lead to a stable auto-inhibitory complex that can be purified for further studies.

Jane's need for distance tends to dominate her desire for closeness, and this internal conflict is reproduced externally in her relationship with Rochester, with Rochester's desire for physical proximity conflicting with Jane's desire for distance. These internal and external power struggles create a healthy sense of tension necessary both to Jane, and to her relationship with Rochester because it prevents either of them from being fully satisfied, and ensures that both remain in a perpetual state of self-inflicted suffering. The suffering these characters impose on themselves and each other is necessary for the preservation of desires, which would be destroyed by fulfillment. Through my reading of the novel we gain a greater understanding of how the pain of unfulfilled desires becomes synonymous with pleasure, and the beneficial role pain, tension and unfulfilled desires plays in the text.

The purpose of this project was to demonstrate the flaws in existing methods of measuring public opinion and to determine the correlation between the age of an offender and the sentencing recommendation received. Show moreIn 2005, the United States Supreme Court found the execution of juvenile offenders to be unconstitutional, based in part on a "national consensus" against the death penalty for juveniles. The purpose of this project was to demonstrate the flaws in existing methods of measuring public opinion and to determine the correlation between the age of an offender and the sentencing recommendation received. It was the hypothesis of my project that there would not be a statistically significant correlation between the offender's age and sentencing recommendation. To test this hypothesis, I asked 156 Florida Atlantic University students. This method of analysis was considered the best way to accurately determine public sentiment towards the juvenile death penalty, after a thorough review of existing methods revealed significant flaws. My results suggest that there is no correlation between the age of an offender and sentencing recommendations.

Blurring the lines between fantasy and reality, these displays --scenes of ghostly hauntings, bloody violence, and excessive emotion--create a desirable uncertainty about the limits of life and love in this novel. This uncertainty constitutes an escape from and an alternative to the conventional romantic relationship prescribed by social and narrative standards in which two people fall in love, get married, have children and die. In my thesis, I argue that the revelation of this desired uncertainty is made possible by Brontèe's use of Gothic devices and could not have been as successfully achieved by any other literary mode.

Show moreThis thesis considers the role and function of God the Son within the anti-Trinitarian system John Milton envisions in his epic poem, Paradise Lost. In the poem, God the Father frequently acts independently of His Son, and the divine attributes that traditionally describe the Son, including His timelessness, no longer apply to the character Milton constructs. Despite this apparent degradation, Milton still elevates the Son and considers Him to be a character worthy of our respect. To account for this seeming paradox, I propose a reading of Paradise Lost that does not dismiss Milton's heretical belief in a subjected Son, but rather uses it as a way envision a new form of power. To do so, I compare the relationship between God the Father and His Son in terms of light and sun imagery, to demonstrate how power is divided and distributed between the two, according to the scientific principles of Milton's day. In addition, I consider how Michel Foucault's concept of the docile body both applies to the Son and explains His deference to the Father. Through these analyses, I hope to demonstrate that the Son's power exists as the result of properly exercising His free will, a will that would not have been His own had He been one with His Father.

Regardless of Bacon's preaching that the progression of scientific knowledge brings one closer to God, in fact human dominance over the natural world undermines religious faith. In particular, Bacon's own interest in early English gardening is used to illustrate the degradation of religious mysticism due to the rise of scientific discovery. Historically, Bacon has been credited as the catalyst of the scientific revolution due to his proposition of a new system of scientific inquiry based on empirical and inductive principles and the development of new arts and inventions. However, Bacon's embrace of the physical sciences and support for human advancement contradicts his alignment with religion. This tension is at the heart of the paper.
