Show moreWe report here the development of an efficient method for the conversion of a variety of conjugated alkynyl esters to alpha-substituted conjugated allenyl esters (racemic) through the use of strong amide bases. Substantially improved yields over typical enolate formation conditions were observed with the use of two equivalents of lithium diisopropylamide. Trapping studies indicate that the second equivalent of base likely leads to the dianion intermediate which upon addition of methyl iodide and trimethylsilyl chloride gives mixtures of alpha-substituted conjugated allenyl and beta,gamma-alkynyl deconjugated esters. Further optimization revealed that additive salts such as LiCl lead primarily to the allenyl product while the use of HMPA as a cosolvent gives the beta,gamma-alkynyl deconjugated alkylation product. The role of base, base concentration and electrophile on product yield and selectivity is also discussed. The optimized conditions resulting from our study of this reaction establishes this method as a viable synthetic tool for the production of alpha-substituted allenyl esters. A new method to prepare racemic alpha-tributylstannyl-alpha-allenyl esters from precursor conjugated alkynyl esters in good yields is reported. Key features of the optimized reaction conditions include the use of multiple equiv. of lithium diisopropylamide in THF with LiCl as a key additive. The allenyl tin reagent reacts with aldehydes in the presence of Lewis acid to give homopropargyl alcohol or allenyl alcohol products depending on the reaction conditions. The addition of boron trifluoride etherate to a solution of the allenyltin and aldehyde gives the homopropargyl alcohol. However, allenyl alcohol products are obtained exclusively with the addition of tin tetrachloride to the allenyl stannane rapidly followed by the addition of aldehyde. The effect of other Lewis acids and reaction variables such as solvent and temperature on the yield and relative stereochemistry of the products will also be described. The Mitsunobu coupling reaction of sterically hindered phenols and alcohols is greatly accelerated by the use of high reaction concentration in combination with sonication. The rate acceleration effect observed with our protocol may simply be based on the high concentration with the sonic waves providing efficient mixing of the highly viscous reaction mixture and generating localized hot spots, or in part, the result of an enhancement of a radical reaction pathway.

Show moreThe pseudopterosins and seco-pseudopterosins are diterpene glycosides isolated from the marine soft coral, Pseudopterogorgia elisabethae . These compounds exhibit anti-inflammatory and analgesic activity greater than the industry standard, indomethacin. The overall goal of this research was to complete biosynthetic studies and enzymology related to the development of a biotechnological production method of the pseudopterosins and seco-pseudopterosins. We aimed to examine early steps in the biosynthetic pathway in order to gain detailed knowledge of the biosynthesis and enzymology of the system. Prior to examination of the biosynthetic pathway leading to the pseudopterosins, we developed both in vivo and in vitro systems to test putative precursors and demonstrated that pseudopterosin A is a precursor to pseudopterosins B--D. We also identified and confirmed the intermediacy of the pseudopterosin diterpene cyclase product using a radioactivity-guided isolation. The bicyclic structure of the diterpene cyclase product suggests that the pseudopterosins and seco-pseudopterosins are derived from this common bicyclic intermediate. The isolation of the pseudopterosin diterpene cyclase product provided us with an assay for the purification of the enzyme involved in its production. We identified the diterpene cyclase in aqueous extracts of P. elisabethae and plan to utilize the amino acid sequence for identification of the gene. This represents the first isolation of a biosynthetic enzyme from a marine coral. Moreover, various characterization studies indicated that the enzyme displays certain similar characteristics to other terpenoid cyclases isolated from terrestrial sources. In previous investigations, P. elisabethae was found only in the Bahamian and West Indian region, but we discovered P. elisabethae in the Florida Keys. Furthermore, due to the distinctive chemistry in the Florida Keys P. elisabethae, plausible early biosynthetic intermediates were isolated that are not present in the Bahamian population. We evaluated these compounds as intermediates in the biosynthesis of the pseudopterosins. The data obtained further supports the assumption of a common biosynthetic origin of the pseudopterosins and seco-pseudopterosins.

Previous crustacean chemoreceptor thresholds are too high because of the adaping effects of high stimulus intensities and the order effects of stimulus presentation sequence used in threshold experiments now in the literature. The present study uses an experimental design geared to produce the lowest adaptation possible and shows that single unit chemoreceptor activity occurs at lower concentrations than any previously documented. The differences between lateral and medial chemoreceptor and the variability of the antennular chemoreceptor population are also treated in the present study. It is concluded that threshold differences do not account for the differential behavioral effect of lateral vs medial antennal filament ablation, and that the observed response variability is inherent in receptor response and important to receptor function.

The cyclization proceeds within 30 seconds at room temperature under the action of potassium tert-butoxide. This work stands apart from the current literature due to the absence of transition metal catalysts and/or harsh reaction conditions. A thorough mechanistic investigation is undertaken to better understand the nature of this unprecedented reaction.

Show moreOver the past twenty years, there has been a significant increase of non-governmental organizations (NGOs) participating in development activities. However, there does not exist in the literature clear and agreed upon measures of performance of successful or high performing NGOs. This dissertation examines three organizational factors affecting NGO performance: characteristics, strategies, and functions. It employs a Delphi Method and a mail survey of 399 U.S.-based NGOs registered with the United States Agency for International Development as of October 1, 1996. Organizational variables, such as decision-making, organizational structure, span of control and hierarchy, communications, types of interactions, program areas, intervening strategies, and diversified funding are identified and rated as highly relevant to NGO performance by the executive officials of the responding NGOs. The study finds that: (1) specific variables related to the organizational characteristics of NGOs were perceived as more important to high performance than the variables related to the strategies and functions of NGOs; (2) responding NGO executives show a positive orientation (perhaps bias) towards both self-assessment of the level of performance of their NGOs and assessment of the importance of organizational variables identified by the Delphi group and the literature; (3) most executives considered their organizations as high performers, but the relationship between positive self-assessment of performance by NGO executives and the variables that define characteristics, strategies, and functions was found not to be statistically significant; (4) there is not a typical organizational pattern by which NGOs can be described because of their diverse and collaborative operational arrangements. This study provides to the field of public administration, organizational studies, public policy, and development administration a better understanding of organizational variables considered important to NGOs' performance from the viewpoint of NGO executives. It employs a methodology not typically associated with public administration research, and its findings take us one step further in the direction of explaining key organizational factors influencing high performance of NGOs and the variables that define these factors.

These differences are governed by geographic origin and period of time of construction. The purpose of this study was to gather data on organs of various periods of time, size and type of construction. The results of this investigation further point out the differences between the classic, or Baroque organ as favored in Germany and the organ in Austria. These differences are the result in no small measure of the different use the organ had in Protestant Germany and Roman Catholic Austria. The method of research consisted of a personal inspection by the writer of some thirty-six organs while on a trip through Germany and Austria in 1970. The results are described in this study.

Percent coverage for each species was estimated using 60 points randomly positioned over the plate area. Physico-chemical conditions at stations 1 and 2 were relatively stable, whereas station 3 was subjected to wide variations in salinity and, to a lesser extent, dissolved oxygen. Plates at stations 1 and 2 consistently collected more species than those at station 3. Peak settling and growth at all stations occurred in the spring and early summer. Three species settled only on plates that had been previously colonized. Even though gelcoat-covered fiber glass was designed to discourage fouling, it proved to be a satisfactory substrate for a thriving community.

Show moreIn the present dissertation, we discuss the development of a stereoselective method for the production of phosphorus compounds that utilizes a phospha-Michael addition reaction. Separately, the design and synthesis of compounds that contain an all-carbon bridged bicyclic scaffold is reported; these compounds were used in initial SAR studies in different in vivo models. In Chapter one is presented a mechanistic framework to develop a highly diastereoselective method catalyzed by phase transfer chemistry leading to phosphinate compounds. In this method, phosphinate nucleophiles were added to various alkenyl ketones as Michael acceptors using crown ethers as phase transfer agents to obtain highly diastereoselective products with the generation of a carbon-based quaternary centers. A closed transition state mechanism is proposed to describe the diastereoselectivity observed in the reactions that is consistent with product outcome as established by X-ray crystallography. Analysis using the 31P NMR technique is also reported to ascertain the diastereomeric ratios in product formation. Using products obtained with the newly developed method, we disclose for the first time a novel phospha-heterocycle with high control of stereochemistry. Relative stereochemistry of the phosphorus containing heterocycle was reported using 2D NMR analysis. In Chapter two focus is placed on the use of acrylates as Michael acceptors in both the diastereoselective and enantioselective studies of phospha-Michael addition. In the asymmetric method development, screening of various chiral catalysts and development of HPLC method to quantify the enantiopurity of products obtained under reaction conditions are reported. The role of crown ether catalysts towards diastereoselectivity is reported.

Barra and Grossman's criticism of Keynes for predicting a negatively sloped labor demand function was analyzed using regression analysis to estimate the function. Barro and Grossman's criticism was found to be based on insufficient evidence, and evidence presented in this thesis supported the Keynesian position.

Show moreThere is a growing interest and attention for organizations to move from a shareholder capitalist to stakeholder capitalist business model, in which delivering environmental, social, and governance (ESG) performance is becoming more pronounced. Because of this transition, there is growing pressure for managers at all levels to act in sustainable, socially responsible, and ethical (SRE) ways; yet the current management models are based on maximizing economic value and scientific management. Therefore, the attempts to try and manage 21st century organizations with 20th century management practices and the aspirations of responsible management education have not demonstrated the desired effect (Laasch & Gherardi, 2019), as seen by continued business failures. It is not yet understood whether it is possible for the everyday manager to effectively navigate the tensions and contradictions of trying to deliver economic value to shareholders, social and ecological value to other stakeholders, while simultaneously attending to regulatory policies and, in the process, avoiding burnout for overtasked managers. Conducted in a debt remediation call center in the Dominican Republic, this study used an eclectic design integrating abductive analysis with a cultural-historical activity theory methodology to explore responsible management practices at the lowest levels of management. The findings describe responsible managers as pressure moderators enacting four primary elements: 1) embracing a pluralistic logic of responsibility, 2) accepting tensions with resilience, 3) engaging practices dynamically to moderate pressures, and 4) learning primarily on the job amid real work. In this organization, moderating the pressures of emotional labor was the primary object of their attention.
