Show moreThe largest barrier to treatment of HIV-1 infection is the establishment of a viral reservoir constituted mostly by quiescent latently infected CD4+ T cells. This reservoir is formed through two processes: i) the infection of resting CD4+ T cells; both naïve and memory, ii) the infection of activated CD4+ T cells which then become quiescent infected cells. One goal of this project was to understand the gene expression changes occurring in naïve CD4+ T cells following activation and subsequent HIV-1 infection and how this may contribute to the establishment of a latent infection in these cells. Utilizing RNA-Seq and a series of validation assays we have identified several genes which are regulated in opposite directions during activation versus infection which we termed DEOC genes. The DEOC genes include a group of physically- and functionally-associated proteins which are key regulators of T cell activation, the cell cycle, cellular proliferation, and cellular quiescence, suggesting that modulation of these DEOC genes may help transition the infected-activated cell from an activated state to a quiescent/resting state to induce a latent infection.

The amplitude (in muV) and latency (in ms) of the P3 component of the auditory ERP were used in the present study to investigate differences between HIV-asymptomatic, HIV-symptomatic, and HIV-negative control adult (18--35 years old) women under three different task conditions. Results indicated that P3 amplitude measures were consistently greater in controls than in the asymptomatic and symptomatic groups for all task conditions and at all electrode locations, suggesting that the available level of attentional resources are reduced similarly in both symptomatics and asymptomatics. P3 latency measures revealed that processing speed by asymptomatics is more similar to controls than it is to symptomatics, indicating that speed of processing is compromised in symptomatics, and possibly, to a lesser degree in asymptomatics.

The objective of this research is to discover improved methods of capturing and counting CD4+T, in a portable assay to aid in the detection of HIV or other diseases that are informed by cell identification and count. The research divides into 4 major objectives: Design an improved portable, microchip. to isolate cells in a timely manner. Explore, design and prove the optical technology that provides large field-of-view and enables imaging large surface area simultaneously so that a sufficient sample can be collected. Test and analyze the microchip and optics to verify the specificity and efficiency of the biological process. Identify and count the cells in an automated manner.

Show morePlasma-based diagnostics are ideal for detecting a variety of diseases because they offer a method of detection that is minimally invasive, readily available, and easy to use for monitoring patients as they progress through a disease or respond to treatment. The only serum marker for PDAC is CA19-9 which lacks specificity, has limited sensitivity, and is unreliable for early detection. It is therefore of great importance to develop a diagnostic that is viable for screening and early detection. Exosomal miRNA were determined via bioinformatics analyses and then examined in PDAC cell lines to identify markers with greatest potential. These markers were then examined in plasma from PDAC patients and control groups. Four markers, miR-93-5p, miR-339-3p, miR-425-5p, and miR-425-3p, emerged as the most viable biomarker panel with the ability to detect PDAC in 100% of the early stages (N=5) compared to CA19-9 which showed increased levels in only one patient with early stage PDAC. Additionally, the diagnostic has a specificity of 80% and a sensitivity of 94.7%, making it comparable to CA19-9, and may even be beneficial for use in conjunction with CA19-9. A plasma-based diagnostic was also developed for multi-strain HIV-1 detection utilizing the loop-mediated isothermal amplification (LAMP) method. LAMP primers were developed against the integrase and vpr regions of the HIV-1 genome. They were tested first in cultured HIV samples and then examined for their ability to amplify HIV-1 subtypes A-G. The integrase primer set provided a reliable means of diagnosing all 55 strains and isolates in under 30 minutes, whereas vpr was inconsistent and exhibited high variability in detecting the HIV subtypes. Our limit of detection for B-subtype with integrase was 30 viral copies/reaction. This could provide the basis for a novel, point-of-care diagnostic for use in underdeveloped regions.

Self-care consisted of daily repetition of an inspirational passage and mantram, which served as the interventions for the primary study. The primary researchers noted that ASCAS scores rose significantly after the intervention. Thus, secondary analysis focused on the relationships of the basic conditioning factors (BCFs) to the total scores on the Adult Self-Care Agency Scale (ASCAS) before and after the self-care intervention. Secondary analysis revealed that diet alone, from among the BCFs, differentiated the sample of HIV-infected or family members of infected persons according to ASCAS scores at study entry and exit.

Show moreThis thesis aims to address the challenges of the development of cost-effective and rapid assays for the accurate counting of CD4+ T cells and quantification of HIV-1 viral load for resource-constrained settings. The lack of such assays has severely affected people living in disease prevalent areas. CD4+ T cells count information plays a vital role in the effective management of HIV-1 disease. Here, we present a flow-free magnetic actuation platform that uses antibody-coated magnetic beads to efficiently capture CD4+ T cells from a 30 μL drop of whole blood. On-chip cell lysate electrical impedance spectroscopy has been utilized to quantify the isolated CD4 cells. The developed assay has a limit of detection of 25 cells per μL and provides accurate CD4 counts in the range of 25–800 cells per μL. The whole immunoassay along with the enumeration process is very rapid and provides CD4 quantification results within 5 min time frame. The assay does not require off-chip sample preparation steps and minimizes human involvement to a greater extent. The developed impedance-based immunoassay has the potential to significantly improve the CD4 enumeration process especially for POC settings.

During the visual ERP sequence an interaction between valence and serostatus was found at the Pz location, F(1,24) = 18.99, p<.001, meaning the late positive potential (LPP) was not modulated between viewing neutral and negative images in HIV-positive women. These findings suggest that the manifestation of HIV in women may alter the neural processing of emotions, though not to the detriment of a subsequent cognitive task.

Thus, secondary analysis focused on the relationships of the basic conditioning factors (BCF) to the global and factor scores on the BSI before and after the intervention of meditation. Secondary analysis revealed a difference in scores between entry and exit on the Global Severity Index (GSI) of the BSI in reference to the basic conditioning factor of age. By the end of the 10 weeks of meditation, age was no longer a factor in causing GSI scores to increase.

SRSF1 plays a prominent role in oncogenesis, immune function, and response to several physiological stimuli. To date, the role of SRSF1 as a regulator of mRNA splicing has been largely considered the main mechanism driving its biological functions and its role in disease. We have now characterized SRSF1’s role in Human Immunodeficiency Virus Type I (HIV-1) transcription. SRSF1 interacts with the 7SK small nuclear ribonucleoprotein (snRNP) to mobilize and activate the positive transcription-elongation factor (P-TEFb), which is then positioned on the HIV-1 promoter to increase the processivity of RNA polymerase II (RNAPolII) and promote the release of the negative regulators of transcription DSIF/NELF. Next, we defined the role of SRSF1 in the transcription of cellular genes utilizing an RNA sequencing (RNASeq) time course approach was used to detect changes in the transcriptome in response to SRSF1 overexpression. RNASeq data analysis revealed a subset of genes that were upregulated in response to SRSF1 overexpression. Nuclear run-on and qPCR assays experimentally validated 28 of these genes.

In order for effective vaccine strategies and modalities to be developed, these earliest immunological events must be fully assessed in a non-biased manner. Nonhuman primates (NHP), specifically Rhesus macaques (RM), serve as a model to investigate the immunological landscape immediately post-challenge and to define the spatiotemporal path of simian immunodeficiency virus (SIV). SIV infection of RM serves as a model of human HIV infection as it recapitulates many of the virological, immunological, and pathological features of HIV infection in the human host. In this thesis I will test the hypothesis whether transcriptional analysis will allow a sensitive measure of the early innate immune responses that accompany detection of the SIV virus in the periphery. I have determined that an early inflammatory profile arises early in tissues proximal to the challenge site that precedes widespread immune activation and the systemic antiviral interferon response. This study defines in detail the spatiotemporal relationship between virus and host immune response and may be a valuable resource in guiding future vaccine design strategies.

Show moreThe complex problems of the twenty-first century cannot be effectively addressed with twentieth century top-down bureaucratic governance alone because of limited stakeholder participation in collective decision making and/or implementation. The somewhat limited stakeholder participation in the policy process, especially, that of target populations, can impact generating viable solutions to complex problems. Collaborative governance has emerged as a promising alternative to traditional management in addressing contemporary complex problems. Collaborative governance is thus a type of governance that promotes joint participation of state and non-state stakeholders in decision making and/or implementation by using agreed upon processes of engagement to collectively address problems. Such governance is especially beneficial for addressing the challenges posed by the human immunodeficiency virus (HIV) and acquired immunodeficiency syndrome (AIDS). This dissertation uses the collaborative governance concept to explicate the efforts of HIV Health Services Planning Councils in Broward County and Palm Beach County of South Florida with the purpose of accentuating how collaborative governance works in providing various categories of services to meet the needs of people with AIDS (PWAs), as part of the implementation of the Ryan White CARE Act. The study focuses on critical variables of collaborative governance such as facilitative leadership and institutional design, the collaborative process variables such as trust building, commitment to the process and shared understanding, and outputs such as allocation priorities of the Councils. This study employed mixed methods in collecting data from various relevant sources., The combined findings from surveys, interviews, observations and document reviews were essential to knowledge and understanding of collaborative governance of the respective Councils. The results revealed more similarities than differences between the Councils in their efforts towards addressing the HIV/AIDS problem. The Councils are similar in all variables of collaborative governance with few exceptions relative to facilitative leadership and institutional design. The differences were relative to membership, number of committees, amount allocated for various service categories and number of clients served. Furthermore, the Councils were slightly different in reaching consensus on subjects of deliberation. On the average, Palm Beach County's Council make decisions by consensus relatively easily when compared with the Broward County's Council. Nonetheless, both Councils are consensus-oriented and strive to make decision by consensus as evidenced by unanimous votes or simple majority votes on various subjects of deliberation. In addition, representation and participation of target populations in collaborative governance have contributed to the empowerment of those target populations. The study contributes to the literature by developing a conceptual model for HIV/AIDS collaborative governance for producing outputs to help meet needs of target populations. Also, the study contributes to collaborative governance theorizing by drawing a nexus between variables of collaborative governance and by complementing the existing non-linear perspective of collaborative process for collective problem solving.
