Show moreThe ubiquitin ligase Highwire is responsible for cell-autonomously promoting synapse formation in the Drosophila Giant Fiber system. highwire mutants show defects in synaptic function and extra branching at the axon terminal, corresponding to transient branching that occur in the course of giant synapse formation during metamorphosis. The MAP kinase pathway, including Wallenda and JNK/Basket, plus the transcription factor Jun, act to suppress synaptic function and axon pruning in a dosage sensitive manner, suggesting different molecular mechanisms downstream of the MAP kinase pathway govern function and pruning. A novel role for Highwire is revealed, regulating the giant fiber axon’s ability to respond to external cues regulated by Fos. When expression of the transcription factor Fos is disrupted in the post-synaptic TTMn or surrounding midline glia of highwire mutants, the giant fiber axons show a marked increase in axon overgrowth and midline crossing. However, synaptic function is rescued by the cell nonautonomous manipulation of Fos, indicating distinct mechanisms downstream of Highwire regulating synaptic function and axon morphology.

As observed in Escherichia coli RNase R (EcR) studies, MgR degrades poly(A), rRNA, and oligoribonucleotides in 3'--->5' direction, though its substrate specificity and optimal activity requirements vary. Interestingly, MgR is sensitive to 2-O-methylation stopping downstream of such modifications in native rRNA and synthetic oligoribonucleotides. MgR removes the 3' trailer sequence from a tRNA precursor of M. genitalium and generates products equal to the mature tRNA, demonstrating a role of MgR in tRNA maturation. The 3' terminal CCA sequence and the acceptor stem of tRNA play a role in determining the formation of such products by MgR. These results suggest multiple functions of RNase R in RNA metabolism in Mycoplasma.

Despite the strides that have been made in ocean exploration, the marine biome has still been only barely explored. Much of the impetus for the isolation and identification of chemical entities from marine organisms has been related to their potential as medicines. For a number of reasons, including synthetic efficiency, this has meant that many marine compounds isolated and studied have been small molecules. In recent years, large strides have been made in techniques for the isolation and identification of biological macromolecules, especially proteins. The understanding of these molecules, and their relationship to the biochemical processes of the marine organisms from which they have been isolated, is important not only because of the potential for this information to help in the synthesis of medicines, but because it may help in the recognition of processes that affect the very viability of marine organisms increasingly exposed to anthropogenic threats to their environment. This work consists of four related studies involving the development of methods for the separation and identification of proteins from a number or gorgon ian species. Chromatographic and gel based methods were used to isolate an elisabethatriene synthase (ELS) which shows promise as a biotechnological tool for the production or pseudopterosins. A number of gorgonians were screened for the presence of proteins that might have antimicrobial properties, and several organisms were identified that might be of interest in this context. Two-dimensional gel electrophoresis methods were then developed to allow the construction of gel maps for the azooxanthellate gorgonian Leptogorgia minimata and the xoozanthellae gorgonian Pseudopterogorgia e/isahethae, the separated proteins were digested and analyzed by LC/MSMS. and the information generated was used to examine the proteome of the organism for functional and phylogenetic relationships. Recently, the cardiac troponin T (cTnT) gene, along with three additional shorter isoforms of the gene, were cloned from normal embryonic hearts. These isoforms are believed to be the alternately spliced forms of the full length gene. One of the isoforms cloned is missing a cardiac-specific exon. Real-time PCR reveals that homozygous recessive mutant embryos (c/c) exhibit a lower transcription level of the cTnT gene than wildtype animals (+/+ or +/c). Expression levels of each of the isoforms are compared in normal and mutant hearts using quantitative real-time PCR.

Real time RT-PCR reveals that mutant hearts express the same level mRNA of the alpha-tropomyosin and TM4 type tropomyosin (ATmC-3) gene as normal embryonic hearts. These genes show no differences with regard to the splicing patterns of normal and mutant. Using protease inhibitor LLnL and E-64d treatments and two-dimensional Western blots of normal and mutant hearts, it is found that mutant hearts express all tropomyosin protein isoforms as normal hearts but protein expression are at low levels. These studies suggest that there is a failure in the translational or posttranslational control mechanisms for tropomyosin protein synthesis in cardiac mutant axolotl hearts during development.

To study the transcriptional and post-transcriptional mechanisms regulating gene expression, the Bcl-x gene has been utilized. A complex mini-gene construct has been create in order to monitor the effects that promoter sequences, 5'UTR and 3'UTR's have on mRNA splicing, RNA export, stability and translation. Abundant evidence exists indicating that RNA processing events such as transcription, splicing and export are coupled, yet the mechanisms and factors involved in regulating these processes are poorly understood. The mini-gene is identical to the endogenous gene with the exception of a deletion to the 50Kb intron and the addition of a tag to differentiate the mini-gene product from the endogenous mRNA and protein. This novel system allows for the study of transcriptional and post-transcriptional mechanisms regulating gene expression from RNA biogenesis on to the protein level.

Show moreLysine-rich KED was previously identified from wounded tobacco (Nicotiana tabacum) leaves before the alignment of protein sequences between NtKED (Nicotiana tabacum KED) and SlKED (Solanum lycopersicum KED) were discovered to display 55.1% identity. Using previously generated SlKED knockout plants by CRISPR/Cas9, we performed biological assays, to investigate the role of KED in wound response to biotic and abiotic stress. Previous studies implied that the KED gene functions as a role in the wound-induced mechanism, as well as suggested that it may also function in the plant defense system against biotic stress and insect herbivory. The results from bioassays using tobacco hornworm (Manduca sexta) have proven inconclusive thus far. Expression of KED is induced not only by mechanical wounding but also by touching such as brushing the leaves, indicating that this gene is sensitive to subtle environmental signal and may be involved in defense response against abiotic stress. To further investigate the KED gene’s role in the plant defense system, biological assays using both specialist and generalist herbivores, transcription analysis using various phytohormone mutant plants, and Evans blue cellular damage assays were performed. Our findings imply that the KED gene does not seem to have a long-term effect on insect herbivory but may have a shortterm anti-feeding effect against insect herbivores. Results from the Evans blue membrane damage assay indicate the KED gene may provide some benefit to mechanically damaged plants in a short-term period post-wounding of leaf tissues. Using the SlKED knockout as genetic tool, we conclude that this gene does not confer resistance to insect herbivores over a long-term but seems to provide a beneficial defense response in the short-term. Our membrane damage assay results also imply that this gene may be involved in membrane stabilization and repair of cellular damage after mechanical wounding.

Bioinformatic studies indicate this is due to overlapping functions remaining after a recent evolutionary divergence. I set out to use in vitro-binding techniques to identify the characteristics of the ZAD family and test this theory. I have constructed glutathione S-transferase (GST)-ZAD domain chimeric proteins for use in pull down protein binding assays,and GST-Zinc finger (ZnF) array domain chimera for electrophoretic mobility shift assays (EMSA). Protein binding assays indicated two putative conserved interactors, similar to the analogous KRAB system in mammals. ... Competitive bindings were carried out to show a specificity of binding conferred by the identified conserved positions. While the consensus binding sites show relatively few similarities, the predicted target genes identified by the consensus binding sites show significant overlap. The nature of this overlap conforms to the known characteristics of the ZAD family but points to a more positive selection to maintain conservation of function.

Fifteen subjects (8 obese and 7 normal-weight) were recruited to participate in an acute HIIE protocol. Our results demonstrated a significant elevation across time in plasma cfDNA and IL-6 immediately following acute HIIE, with no difference between obese and normal-weight subjects. Furthermore, cfDNA was not correlated with IL-6 in response to acute HIIE in either group. These findings indicate that the obese state does not further exacerbate the release of acute HIIE-induced inflammatory mediators (cfDNA and IL-6), which suggests that HIIE training may serve as a time-effective exercise strategy to improve obesity-associated inflammation.

LOLA is a transcription repressor and can generate 17 DNA - binding isoforms, through alternative splicing, each containing distinct zinc-finger proteins. This unique DNAbinding binding sequence to which LOLA-ZFP binds has been determined for four of the lola isoforms F, J, P and K. Also, bioinformatics' tool approach has been taken to identify the target genes that are regulated by these four LOLA splice variants. Future work will be done for the five other LOLA isoforms to categorize their putative DNA-binding sequences and subsequently their protein interactions.

Although, approximately 0.5% of the population suffers from SS, there is a female predominance of 9:1 compared with males. Most women with SS are postmenopausal; however, not all women who are post-menopausal develop SS. Therefore, we postulate that a decrease in the circulating levels of hormones creates an environment favorable to the development of SS in a predisposed genetic background. In order to carry out our studies, we used the NOD.B10.H2b mouse model of SS, and ovariectomized (OVX) them as a model for the post-menopausal condition. We removed the lacrimal glands and measured the gene expression and protein levels of several cytokines and chemokines known to be upregulated in patients with SS such as : lL-1B, IL-10, INF-y, TNFa, CCL9 and CXCL13., We also stained for markers of B cells (B220+) and T cells (CD4+ and CD8+), and counted positively stained cleaved caspase-3 cells as an indication of apoptosis. These experiments were done 3, 7 and 21 days post-OVX and compared to sham operated animals. In order to determine whether the changes observed with OVX were triggered mainly by a genetic pre-disposition, a non-prediposed OVX and sham operated mouse (C57BL/10) was used as control. We found that gene expression of IL-1B, IL-10 and IF-y were upregulated in the lacrimal glands of the OVX NOD.B10.H2b mice at 3 days post-OVX compared with sham operated animals. Gene expression of IL-1B, IL-10, IFN-y, TNF-a, CCL9 and CXCL13, and protein levels of IL-1B, IL-10 and CCL9 were upregulated in the OVX NOD.B10.H2b mice at 7 days post-OVX compared to sham operated animals., Also, at 7 days, an increase in B220+ B cells and an increase in cleaved caspase-3 were also observed in the OVX NOD.B10.H2b mice lacrimal glands compared to sham operated animals. At 21 days, protein levels of IL-10 were also highly upregulated in the OVX NOD.B10.H2b mice, together with an increase of B220+ B cells, a slight increase in the CD4/CD8 ratio and an increase on the number of caspase-3 positive cells. No changes were observed in any of the above parameters measured in the OVX C57BL/10 mice compared to the sham operated group, supporting our hypothesis that both, genetics and a decrease in the levels of hormones are necessary for SS to occur.

Here I examine the roles of Highwire at a central synapse in the adult Drosophila Giant Fiber System (GFS). Highwire is indeed necessary for proper axonal growth as well as synaptic transmission in the GFS. Differences arise between the central synapse and the neuromuscular junction (NMJ), where highwire was initially characterized : expresion from the postsynaptic cell can rescue highwire synaptic defects, which is not seen at the NMJ. In addition, a MAP kinase signaling pathway regulated by highwire at the NMJ has differing roles at a central synapse. Wallenda MAPK can rescue not only the highwire anatomical phenotype but also the defects seen in transmission. Another distinction is seen here : loss of function basket and Dfos enhance the highwire anatomical phenotype while expression of dominant negative basket and Dfos suppress the highwire phenotype. As a result we have compared the signaling pathway in flies and worms and found that the NMJ in the two organisms use a parallel pathway while the central synapse uses a distinct pathway.

In previous work from the laboratory using a patient tissue repository, an isoform of this gene, short form (SIM2-s) was shown to be colon cancer specific. Inhibition of SIM2-s expression by antisense technology resulted in cancer-cell specific apoptosis within 24 hours. Microarray-based gene expression profiling of the antisense-treated colon cancer cells provided a fingerprint of genes involving key cell cycle, apoptosis, DNA damage and differentiation genes. Taking hints from the microarray database, experiments were initiated to decipher the molecular mechanism underlying the cancer specific function of the SIM2-s gene. Using an isogenic cell system, apoptosis was found to be dependent on DNA damage and repair gene, GADD45-a. Further, key pathways including p38 MAP kinase (MAPK) and specific caspases were essential for apoptosis. Programmed cell death was not dependant on cell cycle and was preceded by the induction of terminal differentiation. To clarify whether SIM2-s function is a critical determinant of differentiation, stable transfectants of SIM2-s were established in a murine adipocytic cell line (3T3-L 1 ). SIM2-s overexpression caused a pronounced block of differentiation of the pre-adipocytes into mature adipocytes. A study of the differentiation pathway in 3T3-L 1 cells suggested that this block occurs early on in the cascade. These results supported the starting premise that SIM2-s is a critical mediator of cell differentiation. To clarify whether the SIM2-s gene has transforming potential, the SIM2-s gene was overexpressed in the NIH3T3 murine fibroblast cell line. The cells expressing the human SIM2-s gene exhibited shorter doubling time, abrogation of growth serum requirement, greater cell number at saturation density and focus formation. In vivo tumorigenicity assays showed tumor formation with long latency. These results provide strong evidence for the role of SIM2-s gene in tumor cell growth and differentiation, and validate drug therapy use for the gene.

It is currently unknown how extracellular signals regulate autophagy activity within different tissues during these processes and whether autophagy functions cell-autonomously or nonautonomously. We have data that for the first time shows autophagy activity in the neurons and intestinal cells plays a major role in regulating adult lifespan and the longevity conferred by altered IGF signaling and dietary restriction, suggesting autophagy can control these phenotypes cell non-autonomously. We hypothesize that autophagy in the neurons and intestinal cells is an essential cellular process regulated by different signaling pathways to control wild type adult lifespan, IGF mediated longevity and dietary restriction induced longevity. Excitingly we also have found that in animals with reduced IGF signaling autophagy can control longevity in only a small subset of neurons alone. Autophagy in either specific individual chemosensory neurons or a small group of them is completely sufficient to control IGF mediated longevity. This work provides novel insight to the function and regulation of autophagy which will help shed light on understanding this essential process in higher organisms, including mammals.

LRHR phenotype is proposed as an antecedent to the development of stress hyper responsiveness. Results presented here show emergence of antidepressive-like behavior following peripubertal-juvenile exposure to chronic variable physical (CVP) and chronic variable social stress (CVS) in HR rats, and depressive-like behavior following CVP in the LRs. The antidepressive-like behavior in HR rats was accompanied by increased levels of acetylated Histone3 (acH3) and acetylated Histone4 (acH4) at the hippocampal brain-derived neurotrophic factor (BDNF) P2 and P4 promoters respectively. This effect may mediate increased mossy fibre (MF) terminal field size, particularly the suprapyramidal mossy fibre projection volume (SP-MF), in the HR animals following both stress regimens. These findings show that chronic variable stress during adolescence induces individual differences in molecular, neuromorphological and behavioral parameters between LRs and HRs, which provides further evidence that individual differences in stress responsiveness is an important factor in resistance or vulnerability to stress-induced depression and/or anxiety.
