Show moreThe projects described in this dissertation are focused on compounds derived from the marine environment. Chapter 1 gives an introduction to the study of marine natural products to treat human ailments and a thorough review on compounds from lithistid sponges that have been isolated or synthesized since 2000. Chapter 2 describes the isolation and structure elucidation of two sesquiterpene substituted benzoquinone derivatives, petrosiquinones A and B, from a deep-water marine sponge from the Family Petrosiidae. Although initially purified following activity in a (Sb(B-catenin/Tcf4 assay they were later followed using tumor cell line cytotoxicity assays. Petrosiquinone A was the more active of the two compounds with moderate cytotoxicity in the DLD-1, PANC-1, and AsPC-1 cell lines. In Chapter 3, the isolation and structure elucidation of two new marine-derived macrolides, madeirolide A and B, isolated from a deep-water lithistid sponge of the genus Leiodermatium is described., They were isolated using numerous chromatographic techniques and the structures were elucidated on the basis of 1D and 2D NMR spectra coupled with high resolution-mass spectrometry (HR-MS) data. Madeirolide A and B inhibited the growth of the fungal pathogen Candida albicans with minimum inhibitory concentrations (MIC) of 12.5 and 25 (So(Bg/mL, respectively, but were not cytotoxic in tumor cell assays under the conditions tested. Chapter 4 describes work performed to determine the molecular target of lasonolide A using affinity chromatography. The target of lasonolide A is of interest since lasonolide A is known to kill cancer cells in vitro through a unique mechanism., This chapter highlights the research performed to create an affinity matrix with immobilized lasonolide. A target has not been confirmed but there are a number of interesting hits that are being pursued. In Chapter 5, a liquid chromatography-mass spectrometry (LC-MS) screening method was established in order to rapidly identify the metabolites from numerous collections of Lyngbya spp. obtained from Broward and Lee County, Florida sites that may help marine ecologists assess the effects of Lyngbya spp. blooms on the environment. A link between the metabolites produced and nutrients from both the algal tissue and water column was also explored.

Briareolate esters have been shown to possess biological activity and were previously only reported from Tobago. Our group recently isolated briareolate esters from a specimen collected off the coast of Boca Raton, Florida. To determine whether location has an impact on the chemistry produced by the organism, a method to discern between chemotypes was sought. Several techniques including thin layer chromatography (TLC), high performance liquid chromatography (HPLC), nuclear magnetic resonance (NMR), and sclerite analysis were employed, with NMR being the most successful method. By utilizing both 1H and COSY NMR experiments, it is possible to differentiate between the chemotypes of B. asbestinum. Application of this method allowed analysis of chemical variability with respect to location.

Show moreCoral reefs worldwide are threatened by many environmental disturbances including harmful algal blooms (HABs) which have been increasing on Florida coasts over the past decade. Research has mainly focused on HAB identification, percent cover and other effects on the benthos but the relationship of HABs with upper trophic levels has received less attention. To study this relationship, a two-year investigation on a 10-m deep reef off of Hallandale Beach, was conducted. Stationary fish census coupled with benthic transect videos were conducted quarterly between April 2005-July 2007. A significant correlation between Lyngbya sp. blooms and alterations in fish assemblages was observed. Lyngbya had a negative interaction with fish species and abundance; additionally, HAB sample periods were significantly different from low algal abundance periods. Blooms of Dictyota sp. had no measurable relationship with the fish assemblage indicating HABs may have variable impacts on fishes depending on family, species and chemistry of the algae.

Describing these relationships in endangered marine turtle populations and their critical foraging grounds is essential for determining informed management decisions. This study systematically describes the foraging niche of hawksbills Eretmochelys imbricata, and green turtles, Chelonia mydas in Buck Island Reef National Monument, U.S. Virgin Islands, a critical habitat for nesting and foraging. It assesses the relationships within and between the species in terms of overlap, annual and seasonal variation, and life history and feeding strategies within the community. Most importantly it describes these relationships with metrics that can be used in global comparisons or to measure change in local conditions.

The project detailed in this thesis focuses on the extraction of compounds from marine invertebrates as well as defining the absolute configuration for a compound. Utilizing marine invertebrates, the sonications method developed in this thesis provides an alternative approach to rapidly extract compounds for primary screening. This method is viable compared to a traditional overnight extraction method, without suffering compound degredation... Previously, clathric acid was isolated from an unknown Clathria sp. This compound is a bibyblic C-21 terpenoid shown to have mild antimicrobial activity against gram positive bacteria. With only its relative configuration established, additional amounts of clathric acid were required to define the overall absolute configuration. Identifying the Clathria sp. to be Clathria compressa, through spicule analysis, additional sponge tissues were then collected off the coast of Boca Raton, Florida to isolate additional quatities of clathric acid. The absolulte configuration was determined through circular dichroism and the octant rule to establish a final configuration for clathric acid's four carbon stereocenters to be: (3S, 7S, 8R, and 12S).

From January to March 2008, I conducted a small-scale experiment with three captive Scarlet Ibis (Eudocimus ruber) at the Palm Beach Zoo, in West Palm Beach, Florida. Prey density and the presence of conspecifics were manipulated to test for a response by ibis in terms of prey capture rate. Mixed model analysis, when one prey type was available, showed that capture rate increased significantly with prey density and decreased with the presence of conspecifics. When two prey types were available, prey density and the presence of conspecifics had no effect on the capture rates of either prey type. Selectivity analysis revealed that ibis always selected for crayfish in mixed treatments. I hypothesize that the number of prey types available in a treatment played an important role in determining how fish density and the presence of conspecifics affected ibis foraging success.

This complex relationship is described by interpretations of conservation discourse recorded through ethnographic interviews that demonstrate how these perceptions have been influenced by factors such as personal experiences, film and text, and broad changes in the relationship between humans and nature since the early days of the environmental movement. By linking these perceptual changes with changes in American shark conservation policy, this work not only explains a relationship between culture, perception, and policy, but also celebrates the emergence of a multispecies marine community.

Recently, two new briarane diterpenoids, briareolate esters J and K, together with two known briareolate esters have been isolated from a specimen of Briareum asbestinum collected off the coast of Boca Raton, Florida. The method used was a 96-well plate real-time cell electronic sensing (RT-CES) system to discover compounds that impact human embryonic stem cell growth. The compounds were isolated using reversed phase polystyrene divinylbenzene chromatographic support HP20ss followed by normal phased HPLC using a luna silica column. The structures of the compounds were established though the interpretation of spectroscopic data. Activity testing was conducted against hESCs (BG02) with briareolate ester J showing no inhibition activity and briareolate ester K showing mild activity with an EC50 value of 25 (So(BM. These results confirm that the exact confirmation and existence of the (E,Z)-dienone is related to the activity that was observed with the previously isolated briareolate esters L and M.

However, the releases of methane and carbon dioxide gasses from peat soils are currently not well understood, particularly since the timing of the releases are poorly constrained. Furthermore, most research work performed on peatlands has been focused on temperate to sub-arctic peatlands, while recent works have suggested that gas production rates from low-latitude peat soils are higher than those from colder climates. The purpose of the work proposed here is to introduce an autonomous Ground Penetrating Radar (GPR) method for investigating the timing of gas releases from peat soils at the lab scale utilizing samples originating from Maine and the Florida Everglades, and at the field scale in a Maine peatland. Geophysical data are supported by direct gas flux measurements using the flux chamber method enhanced by timelapse photography, and terrestrial LiDAR (TLS) monitoring surface deformation.

Additional statistical analysis was performed to determine which industries within each MSA explain the greatest amount of variance in the night light data. Industrial variables exhibited strong multi-collinearity. It is therefore impossible to determine which industries explain the greatest variance in the night light image data.

Show moreThis study examined and compared the developmental experiences of leaders who have successfully developed their capacities to engage in and maintain a vision and practice for sustainability over time in themselves and others in business, education and community. Through the application of grounded theory as a systematic comparative method for the analysis of qualitative data, the study generated a substantive theory of leadership called regenerative leadership. This leadership approach is especially relevant at a time when humankind is faced with accelerating change and increasing evidence that numerous natural and social systems at the global level have reached or are rapidly approaching points of overshoot and collapse. Among the major findings, the study revealed that the more evolved sustainability leaders are becoming increasingly dissatisfied with the construct of sustainability, and indicate the need for a profound cultural shift towards regenerative human systems. In this framework, regenerative organizations are driven by a sense higher purpose, and leadership is exercised heterarchically. Leaders and followers engage in generative conversations to create desirable futures which are then backcasted to eliminate unanticipated consequences. Throughout, participants emphasized the critical importance of engaging in personal and collective consciousness development or "inner work" in order to make regenerative practices possible.

In a 52-kmP 2 P study area on the east coast of central Florida, the characteristics of hydrologically diverse ditches were quantified, and their use by Wood Storks documented during their non-breeding season. Logistic regression analyses were carried out using the ditch characteristics as independent variables and Wood Stork presence/absence as the dependent variable. This study confirms the use of these marginal wetlands, and identifies the significance of emergent vegetation on the foraging habitat selection of Wood Storks in the dry season.

Surveys were conducted on approximately 30 linear km of reef between 15 and 30 m in depth. Tissue samples were retrieved from 112 turtles for mtDNA haplotype determination. GPS-linked satellite transmitters were deployed on six resident sub-adults, resulting in both minimum convex polygon (MCP) and 95%, 50%, and 25% kernel density estimates (KDE) of home-range size. A foraging ethogram was developed, and sequential analysis performed on thirty videos (141 total minutes) of in-situ foraging behavior. Seventeen total haplotypes were identified in this aggregation, the majority (75%) of which represented rookeries on Mexico’s Yucatan Peninsula. Other sources, from most to least important, include Barbados, Costa Rica, Puerto Rico, Antigua, and the U.S. Virgin Islands.

By integrating kinematic, musculoskeletal, material properties, and compositional analyses across a range of morphologically and phylogenetically diverse batoids, this dissertation (i) demonstrates that punting is not confined to the skates, and (ii) provides reliable anatomical and mechanical predictors of punting ability. Batoids in this study performed true punting (employing only pelvic fins), or augmented punting (employing pectoral and pelvic fins). Despite the additional thrust from the pectoral fins, augmented punters failed to exceed the punting c apabilities of the true punters. True punters' pelvic fins had greater surface area and more specialized and robust musculature compared to the augmented punters' fins. The flexural stiffness of the main skeletal element used in punting, the propterygium, correlated with punting ability (3.37 x 10-5 - 1.80 x 10-4 Nm2). Variation was due to differences in mineral content (24.4-48-9% dry mass), and thus, material stiffness (140-2533 MPa), and second moment of area. The propterygium's radius-to-thickness ratio (mean = 5.52 +-0.441 SE) indicated that the propterygium would support true and augmented punters, but not non-punters, in an aquatic environment. All propterygia would fail on land. Geometric and linear morphometric analyses of 61 batoid pelvic girdles demonstrated that pelvic girdle shape can predict punting and swimming ability and taxonomic attribution to Order., Characteristics of true punters' pelvic girdles, such as laterally facing fin articulations, large surface area formuscle attachment, and tall lateral pelvic processes are similar to characteristics of early sprawled-gait tetrapods' pelvic girdles. This dissertation demonstrates that punting is common in batoids, illustrates the convergent evolution of true punter and early tetrapod pelvic anatomy, and gives possible explanations for the restriction of elasmobranchs to aquatic habitats.

However, the burrows created have major consequences on the mangrove habitat and aerial root inhabitants. It has been suggested that sessile species residing in aerial root communities can either encourage or discourage colonization by S. terebrans. Abiotic factors can affect the distribution and abundance of mangrove forest and are the same factors which determine the composition and abundance of organisms living on the roots. Surveys indicated that burrowing damage was found predominately in the first 20 cm of the root tip. Exploratory Structural Equation Modeling (SEM) was used to test multivariate hypothesized models looking at habitat relationships with S. terebrans in aerial root communities. Temperature and dissolved oxygen were shown to be important drivers in affecting submerged root length of aerial roots. Ultimately, the indirect effects between these parameters proved to be stronger in influencing the barnacle – isopod association, which causes direct negative effects on submerged root length. Colonial tunicates showed weak effects in masking aerial roots from the damaging barnacle – isopod association. Chlorophyll a was used as a proxy for phytoplankton biomass and proved to be less influential than habitat protection for S. terebrans. Results highlight the need for experimentation in addition to modeling in order to determine the mechanisms influencing aerial root community inhabitants and further effects on the habitat.

On a landscape scale, there appeared to be a relationship among resource availability, the temporal scale of the independent variable, and whether the response was similar or different between species. At the patch level, results demonstrated a relationship between resource availability and the spatial scale of the independent variables selected by birds. Species movements were consistent with the differing strategies. This study is the first to make the link between landscape hydrology patterns, prey availability, and responses in wading bird habitat selection at multiple spatial scales.

I have formulated a model that incorporates vertical movement into the Rosezweig-MacArthur (R-M) predator-prey model. The R-M model is stable only at a carrying capacity below a critical value. I found that adding vertical movement stabilizes the system even at a high carrying capacity. By analyzing temporal stability and spatial structure, my results show that vertical movement interacts with carrying capacity to determine patch structure.

Show moreAquatic plants and submerged aquatic vegetation (SAV) are some of the most wide-ranging species and create important habitat for fish and wildlife in many ecosystems, including highly variable coastal ecotones. Mechanistically understanding factors controlling current distributions of these species is critical to project future distribution and abundance under increasing variability and climate change. I used a population-based approach to quantify the effects of spatial and temporal variability on life history transitions of the SAV Ruppia maritima L. (widgeongrass) in the highly dynamic Everglades-Florida Bay ecotone as a model to (1) examine which life history stages were most constrained by these conditions and (2) determine how management can promote life history development to enhance its distribution, an Everglades restoration target. Ruppia maritima life history transitions were quantified in a series of laboratory and field experiments encompassing a ra nge of abiotic and biotic factors known to affect seagrass and SAV (salinity, salinity variability, temperature, light and nutrients and seed bank recruitment and competition). These studies revealed that R. maritima life history varied east to west across the Everglades ecotone, driven by multiple gradients in abiotic factors that constrained different life history transitions in distinct ways. Based on this examination, persistence of SAV populations from dynamic coastal environments is highly dependent on large reproductive events that produce high propagule densities for recruitment. Large productive meadows of SAV also depend on high rates of clonal reproduction where vegetation completely regenerates in a short amount of time. Therefore, in hydrologically variable systems, maintenance or increases in SAV reproduction is required for population persistence through recruitment. However, SAV communities that do not experience high rates of sexual reproduction are dependent on successful seed germination, seedling and adult survival and clonal reproduction for biomass production and maintenance. Seedling survival and to a lesser extent, adult survival, are bottlenecks that can limit life history transitions under highly variable hydrological conditions. To ensure long-term survival in these communities, management activities that increase survival and successful life history development through these critical stages will be beneficial. If not, SAV populations may become highly reduced and ephemeral, providing less productive habitat.

The purpose of this thesis is to map Australian Pine (Casuarina equisetifolia with a 85% or greater accuracy by creating a methodology that uses LIDAR and color infrared ortho-photography and to test it within three different landscape types within Broward County. LIDAR features below a determined height threshold (i.e. Deerpoint 25 ft) were eliminated and recoded to 0 to create Mask 1. NDVI technique separated non-vegetative features from vegetative features to create Mask 2. Mask 1 and Mask 2 were merged and overlaid on the raw LIDAR data set to perform isodata clustering, as well as density slicing to identify mature Australian Pines. Careful delineation of study areas is critical to obtain the highest possible accuracy. Density slicing proved to be a faster and less time consuming technique for achieving 85% level of accuracy than compared to isodata clustering.

Show moreColonization of infauna in a newly constructed mangrove environment at Spanish River Park, Boca Raton, Florida was studied for six months following initial site preparation. Monthly core samples for benthic invertebrates were collected from December 1994 to June 1995. Sampling stations were chosen randomly each month in three zones along two transects across a tidal channel: upper mangrove, lower mangrove, and mangrove/Spartina. Fiddler crab burrows were counted within randomly placed square meter quadrats in each zone. Rapid succession over short-term indicated the constructed environment would eventually become an established, functional mangrove community. Natural infaunal colonization began immediately, and a total of 26 species was collected during the study period. The most abundant species were microdrile oligochaetes, capitellid polychaetes, and dipteran larvae. Fiddler crab holes were first encountered in April 1995 and reached a maximum in June of 42 burrows per m^2. No significant zonation (p > 0.05) was found in the most abundant species, average infaunal density, or fiddler crab holes. Samples from two reference sites showed similar faunas, with the organisms found in adjacent channel areas an influencing factor on species diversity and density.
