Show moreElectrical impedance of cells is a sensitive indicator of changes in cellular structure and biophysical characteristics. Integration of electrical impedance sensing in microfluidics can be a useful tool for characterization of blood cells for their disease state, such as sickle cell disease and malaria. The first part of this dissertation presents application of a microfluidics-based electrical impedance sensor for the study of sickle cell disease. Dynamic cell sickling-unsickling process of blood cells in response to cyclic hypoxia was measured. Strong correlation was found between the electrical impedance data and patients’ hematological parameters such as levels of sickle hemoglobin and fetal hemoglobin. In addition, application of electrical impedance spectroscopy in narrow microfluidic channel was used for label-free flow cytometry and non-invasive assay of single sickle cells under controlled oxygen level. We demonstrate the capability of this new technique in differentiating normal red blood cells from sickle cells, as well as sickled cells from unsickled cells, using normoxic and hypoxic conditions. The second part of this dissertation reports an application of electrical impedance sensing for the study of placental malaria. Testing conditions were optimized so that electrical impedance can be used for real time monitoring of different cellular and molecular level variations in this in vitro model of placental malaria. Impedance characteristics of cell proliferation, syncytial fusion and long-term response of BeWo cells to adhesion of infected erythrocytes were obtained and related to the immunostaining results and inflammatory cytokines measurements. Comparing to the conventional optical microscope-based methods, electrical impedance sensing technique can provide a label-free, real-time monitoring tool to study erythrocytes and cytoadhesion, and can further be extended to other disease models and cell types.

Show morePalliation therapy for dysphagia using esophageal stents is the current treatment of choice for those patients with inoperable esophageal malignancies. However, the stents currently used in the clinical setting, regardless of the type of metal mesh or plastic mesh stents (covered/uncovered), may cause complications, such as tumor ingrowth and stent migration into the stomach. Furthermore, metal mesh stents have limited capacities for loading anti-cancer drugs. To effectively reduce/overcome those complications and enhance the efficacy of drug release, we designed and 3D-printed a tubular, flexible polymer stent with spirals, and then load anti-cancer drug, paclitaxel, on the stent for drug release. Non- spiral 3D-printed tubular and mesh polymer stents served as controls. The self-expansion and anti migration properties, cytotoxicity, drug release profile, and cancer cell inhibition of the 3D-printed stent were fully characterized. Results showed the self-expansion force of the 3D-printed polymer stent with spirals was slightly higher than the stent without spirals. The anti-migration force of the 3D-printed stent with spirals was significantly higher than the anti-migration force of a non-spiral stent. Furthermore, the stent with spirals significantly decreased the migration distance compared to the migration distance of the non-spiral 3D-printed polymer stent. The in vitro cytotoxicity of the new stent was examined through the viability test of human esophagus epithelial cells, and results indicated that the polymer stent does not have any cytotoxicity. The results of in vitro cell viability of esophageal cancer cells further indicated that the paclitaxel in the spiral stent treated esophageal cancer cells much more efficiently than that in the mesh stent. Furthermore, the results of the in vitro drug release profile and drug permeation showed that the dense tubular drug-loaded stent could efficiently be delivered more paclitaxel through the esophageal mucosa/submucosa layers in a unidirectional way than mesh stent that delivered less paclitaxel to the esophageal mucosa/submucosa but more to the lumen. In summary, these results showed that the 3D-printed dense polymer stent with spirals has promising potential to treat esophageal malignancies.

Show moreSeagrass is a key stone component for the Indian River Lagoon (IRL) ecosystem, and therefore it is an important topic for many studies in the lagoon. This study focuses on the effects of seagrass beds on the hydrodynamics in the IRL. A hydrodynamic model based on the Delft3D modeling system has been developed for the southern IRL including the St. Lucie estuary, Ft. Pierce and St. Lucie Inlets, and adjacent coastal waters. The model is driven by freshwater inputs from the watershed, tides, meteorological forcing, and oceanic boundary forcing. The model has been systematically calibrated through a series of numerical experiments for key parameters, particularly the bottom roughness, and configuration including heat flux formulation and bottom bathymetry. The model skills were evaluated with quantitative metrics (point-to-point correlation, root-mean-square difference, and mean bias) to gauge the agreements between model and data for key variables including temperature, salinity, and currents. A three-year (2013-2015) simulation has been performed, and the results have been validated with available data including observations at HBOI Land-Ocean Biogeochemistry Observatory (LOBO) stations and in situ measurements from various sources. The validated model is then used to investigate the effects of 1) model vertical resolution (total number of model vertical layers), 2) spatial variability of surface winds, and 3) seagrass beds on the simulated hydrodynamics. The study focuses on the vicinity of Ft. Pierce Inlet, where significant seagrass coverage can be found. A series of numerical experiments were performed with a combination of different configurations. Overall, the experiment with 2-dimensional (2-D) winds, ten vertical layers and incorporating seagrass provided the most satisfactory outcomes. Overall, both vertical resolution and spatial variability of surface winds affect significantly the model results. In particular, increasing vertical resolution improves model prediction of temperature, salinity and currents. Similarly, the model with 2-D winds yields more realistic results than the model forced by 0-D winds. The seagrass beds have significant effects on the model results, particularly the tidal and sub-tidal currents. In general, model results show that both tidal and sub-tidal currents are much weaker due to increase bottom friction from seagrass. For tidal currents, the strongest impacts lie in the main channel (inter-coastal waterway) and western part of the lagoon, where strong tidal currents can be found. Inclusion of seagrass in the model also improves the simulation of sub-tidal currents. Seagrass beds also affect model temperature and salinity including strengthening vertical stratification. In general, seagrass effects vary over time, particularly tidal cycle with stronger effects seen in flood and ebb tides, and seasonal cycle with stronger effects in the summer than in winter.

Show moreThere have been much technological advances and research in Unmanned Surface Vehicles (USV) as a support and delivery platform for Autonomous/Unmanned Underwater Vehicles (AUV/UUV). Advantages include extending underwater search and survey operations time and reach, improving underwater positioning and mission awareness, in addition to minimizing the costs and risks associated with similar manned vessel operations. The objective of this thesis is to present the design and development a high-level fuzzy logic guidance controller for a WAM-V 14 USV in order to autonomously launch and recover a REMUS 100 AUV. The approach to meeting this objective is to develop ability for the USV to intercept and rendezvous with an AUV that is in transit in order to maximize the probability of a final mobile docking maneuver. Specifically, a fuzzy logic Rendezvous Docking controller has been developed that generates Waypoint-Heading goals for the USV to minimize the cross-track errors between the USV and AUV. A subsequent fuzzy logic Waypoint-Heading controller has been developed to provide the desired heading and speed commands to the low-level controller given the Waypoint-Heading goals. High-level mission control has been extensively simulated using Matlab and partially characterized in real-time during testing. Detailed simulation, experimental results and findings will be reported in this paper.

While there are many water treatment methods, lime softening proves to be one of the more effective approaches as it removes a wide variety of harmful compounds including arsenic, lead, mercury, and cadmium under the correct conditions. The greatest issues with lime softening on a smaller scale include the complexity of the chemistry and need for monitoring. By designing the system for groundwater sources and with a smaller capacity, this thesis hoped to reduce the level of monitoring, chemical expertise, and cost needed for operation. While promising results occurred in the removal of arsenic and total hardness, this project was unable to obtain consistent results and final water samples with pH values between the recommended standard of 6.5 to 8.

Show moreAutomatic target recognition capabilities in autonomous underwater vehicles has been a daunting task, largely due to the noisy nature of sonar imagery and due to the lack of publicly available sonar data. Machine learning techniques have made great strides in tackling this feat, although not much research has been done regarding deep learning techniques for side-scan sonar imagery. Here, a state-of-the-art deep learning object detection method is adapted for side-scan sonar imagery, with results supporting a simple yet robust method to detect objects/anomalies along the seabed. A systematic procedure was employed in transfer learning a pre-trained convolutional neural network in order to learn the pixel-intensity based features of seafloor anomalies in sonar images. Using this process, newly trained convolutional neural network models were produced using relatively small training datasets and tested to show reasonably accurate anomaly detection and classification with little to no false alarms.

Modified boundary layers (MBLs) inspired by the down coat of an owl’s wing has shown to reduce the acoustic effects caused by flow noise. This thesis investigates the mechanisms that modified boundary layers can provide for reducing the surface pressure fluctuations in a boundary layer. This study analyzes various types of MBLs in a wall jet wind tunnel through computational fluid dynamics and numerical surface pressure spectrum predictions. A novel surface pressure fluctuation spectrum model is developed for use in a wall jet boundary layer and demonstrates high accuracy over a range of Reynolds numbers. Non-dimensional parameters which define the MBL’s geometry and flow environment were found to have a key role in optimizing the acoustic performance.

Type C and type F DC-RCP were used. An anodic current density of various magnitudes (0.5 μA/cm2, 1 μA/cm2 and 2.5 μA/cm2) was applied during the corrosion propagation stage. The specimens were placed in high humidity and selected specimens were later covered with wet sand. Selected specimens were terminated for visual examination and gravimetric analysis. Typically, the reinforcement potentials during the accelerated corrosion period were more negative for F specimens compared to C specimens. The C specimens experienced ~2× more corrosion than the F specimens. The accumulated corrosion products did not cause cracks. A method was developed that allows for modest corrosion acceleration during the corrosion propagation stage of DC-RCP.

Show moreThe design and validation of a low-level backstepping controller for speed and heading that is adaptive in speed for a twin-hulled underactuated unmanned surface vessel is presented. Consideration is given to the autonomous launch and recovery of an underwater vehicle in the decision to pursue an adaptive control approach. Basic system identification is conducted and numerical simulation of the vessel is developed and validated. A speed and heading controller derived using the backstepping method and a model reference adaptive controller are developed and ultimately compared through experimental testing against a previously developed control law. Experimental tests show that the adaptive speed control law outperforms the non-adaptive alternatives by as much as 98% in some cases; however heading control is slightly sacrificed when using the adaptive speed approach. It is found that the adaptive control law is the best alternative when drag and mass properties of the vessel are time-varying and uncertain.

Microdroplets have proven effective in biomedical applications such as single-cell analysis, DNA sequencing, protein partitioning and drug delivery. Conventionally, a series of aqueous microdroplets containing biosamples is generated and controlled in an oil environment. One of the critical challenges in this system is that recovery of the aqueous samples from the oil phase is very difficult and often requires expensive and cumbersome post-processing. Also, the low Reynolds (Re) number characteristic of this system results in low throughput of droplet generation. To circumvent challenges and fully utilize microdroplets for practical clinical applications, this research aims to unpack the fundamental physics that governs droplet generation in oil-free systems including an aqueous two-phase system (ATPS) and a high inertial liquid-gas system.

Show moreThe research shows a novel approach for the Magnetic Anomaly Differentiation and Localization Algorithm, which simultaneously localizes multiple magnetic anomalies with weak total field signatures (tens of nT). In particular, it focuses on the case where there are two homogeneous targets with known magnetic moments. This was done by analyzing the magnetic signals and adapting Independent Component Analysis (ICA) and Simulated Annealing (SA) to solve the problem statement. The results show the groundwork for using a combination of fastICA and SA to give localization errors of 3 meters or less per target in simulation and achieved a 58% success rate. Experimental results experienced additional errors due to the effects of magnetic background, unknown magnetic moments, and navigation error. While one target was localized within 3 meters, only the latest experimental run showed the second target approaching the localization specification. This highlighted the need for higher signal-to-noise ratio and equipment with better navigational accuracy. The data analysis was used to provide recommendations on the needed equipment to minimize observed errors and improve algorithm success.

However, SAS processing requires strict constraints on resolvable platform motion. The most common approach to estimate this motion is to use the Redundant Phase Center (RPC) technique. Here the ping interval is set, such that a portion of the sonar array overlaps as the sensor moves forward. The time delay between the pings received on these overlapping elements is estimated using cross-correlation. These time delays are then used to infer the pingto-ping vehicle motion. Given the stochastic nature of the operational environment, some level of decorrelation between these two signals is likely. In this research, two iterative signal decomposition methods well suited for nonlinear and non-stationary signals, are investigated for their potential to improve the Time Delay Estimation (TDE). The first of this type, the Empirical Mode Decomposition (EMD) was introduced by Huang in the seminal paper, The empirical mode decomposition and the Hilbert spectrum for nonlinear and non-stationary time series analysis and is the foundation for the algorithms used in this research. This method decomposes a signal into a finite sequence of simple components termed Intrinsic Mode Functions (IMFs). The Iterative Filter (IF) approach, developed by Lin, Wang and Zhou, builds on the EMD framework. The sonar signals considered in this research are complex baseband signals. Both the IF and EMD algorithms were designed to decompose real signals. However, the IF variant, the Multivariate Fast Iterative Filtering (MFIF) Algorithm, developed by Cicone, and the EMD variant, the Fast and Adaptive Multivariate Empirical Mode Decomposition (FAMVEMD) algorithm, developed by Thirumalaisamy and Ansell, preserve both the magnitude and phase in the decomposition and hence were chosen for this analysis.

Transfer learning methods are used to replace the need of large datasets, by using a pre trained network on the side-scan sonar images. In the present study the implementation of a generative adversarial network is used to generate meaningful sonar imagery from a small dataset. The generated images are then added to the existing dataset to train an image detection and classification algorithm. The study looks to demonstrate that generative images can be used to aid in detecting objects of interest in side-scan sonar imagery.

A series of steps based on increasing complexity are considered sequentially in this study. First the steady flow past the static vehicle, then the vehicle with a fixed orientation advancing in calm water, and finally the vehicle moving with two degrees of freedom (DOF) in calm water as well as head seas. The main objective of the study is to assess the role of general multiphase unsteady Reynolds Averaged Navier Stokes (RANS) as a predictive tool for the hydrodynamic performance of an USV. A parametric analysis of the vehicle performance at different Froude number and wave steepness in shallow waters is conducted. The characteristics of the wave resistance, heaving and pitching motion, wave-hull interactions, and free surface flow patterns are investigated. The study will aid in the design of a robust physics-based control system for the vehicle and provide a tool for prediction of its performance.

Show moreThe influence of voids on the moisture uptake of epoxy has been studied. Specimens with void contents from 0 to about 50% were prepared. Void geometry and content were analyzed using microscopy and density methods. Void containing dry samples were characterized by Differential Scanning Calorimetry and Dynamic-Mechanical Analysis which verified consistency of chemistry of the epoxy network. The moisture uptake of specimens immersed in distilled water at 40 °C was monitored. The rate of absorption and saturation moisture content increased with increasing void content. The moisture uptake of void-free and void containing specimens was non-Fickian. The Langmuir model provided good fits to the experimental results for specimens with low to medium void content, although the moisture uptake of the high void content specimens showed substantial deviations from the Langmuir diffusion model. The moisture diffusivity agreed reasonably with predications from the Maxwell inclusion model over a range of void contents from 0 to 50%. The state of sorbed water was examined using mass balance calculations and DSC analysis. Only 6-8% of the void volume is occupied by water at saturation. Absorbed water may be classified as free and bound water. For void-free specimens, only bound water was found. The medium and high void content specimens contained water in three states: free water, freezable bound water, and non-freezable bound water. The DSC results show that the proportions of free water and freezable bound water increase with increasing void content, while the content of non-freezable bound water decreased. Moisture induced swelling decreased with increasing void content. The swelling is attributed to the content of non-freezable bound water.

A detailed incompressible numerical study of a single stage axial-flow turbine at a low Reynolds number is investigated with the use of multiple eddy-viscosity turbulence models. The study includes epistemic uncertainty quantification as a form of numerical error estimation. The numerical results show good qualitative and quantitative agreement with experimental data. It was found that the shear stress transport (SST) k - ω turbulence model with rotation/curvature correction and inclusion of transition modeling is most capable at predicting the mean velocity distribution, which is further enhanced when the URANS formulation is employed. However, all the cases indicate a large variation in the prediction of the root-mean-squared of the turbulent velocity fluctuations.

Show moreThis work uses computational fluid dynamics to study the flowfield around a hypersonic missile with two lateral jets to provide control in place of control surfaces. The jets exhaust an H2-O2 mixture at Mach number of 2.9 with a jet pressure ratio of roughly 10,500. The jets are staggered axially and circumferentially in such a way to produce pitch and yaw. The flowfield of such a jet configuration is characterized at several angles of attack and the corresponding force coefficients and amplification factors are provided. The freestream air and H2-O2 plume is treated as inert for the majority of the calculations. Special cases are treated with finite rate chemical kinetics and compared to the inert flowfield to ascertain the effects that chemical reactions have on the force coefficients. It was found that the flowfield was only slightly altered from the familiar one jet flowfield when the second jet is active. The flow topology and vortex structures tend to shift towards the second jet but the overall structure remains the same. The normal force amplification factors are close to unity over the range of angle of attack due to the thrust being so high with the two jet configuration having a lower amplification factor compared to firing a single jet. Treating the flowfield as chemically reacting did not affect the force values much: the difference being 0.3% for an angle of attack of 0°.

Show moreThe research presented in this thesis utilizes Blade Element Momentum (BEM) theory with a dynamic wake model to customize the OrcaFlex numeric simulation platform in order to allow modeling of moored Ocean Current Turbines (OCTs). This work merges the advanced cable modeling tools available within OrcaFlex with well documented BEM rotor modeling approach creating a combined tool that was not previously available for predicting the performance of moored ocean current turbines. This tool allows ocean current turbine developers to predict and optimize the performance of their devices and mooring systems before deploying these systems at sea. The BEM rotor model was written in C++ to create a back-end tool that is fed continuously updated data on the OCT’s orientation and velocities as the simulation is running. The custom designed code was written specifically so that it could operate within the OrcaFlex environment. An approach for numerically modeling the entire OCT system is presented, which accounts for the additional degree of freedom (rotor rotational velocity) that is not accounted for in the OrcaFlex equations of motion. The properties of the numerically modeled OCT were then set to match those of a previously numerically modeled Southeast National Marine Renewable Energy Center (SNMREC) OCT system and comparisons were made. Evaluated conditions include: uniform axial and off axis currents, as well as axial and off axis wave fields. For comparison purposes these conditions were applied to a geodetically fixed rotor, showing nearly identical results for the steady conditions but varied, in most cases still acceptable accuracy, for the wave environment. Finally, this entire moored OCT system was evaluated in a dynamic environment to help quantify the expected behavioral response of SNMREC’s turbine under uniform current.

Show moreLaboratory experiments were conducted to observe, document and evaluate the mechanical behavior of Fiber Reinforced Concrete after being submitted to five different environments for 8 months. The specimens were molded and reinforced with synthetic fibers with a composition similar to that used for dry-cast concrete. Four different types of fibers with different composition were used. The fibers were mixed with the concrete to create the samples and the samples were exposed to different environmental conditions. Some of these environments were meant to increase degradation of the interface fiber-concrete to simulate longevity and imitate harsh environments or marine conditions. The environments consisted of: a high humidity locker (laboratory conditions), submerged in the Intracoastal Waterway in a barge (SeaTech), a wet/dry cycle in seawater immersion simulating a splash/tidal zone, low pH wet/dry seawater immersion cycle and samples submerged in calcium hydroxide solution. The latter three were in an elevated temperature tank (87-95°F) to increase degradation process. The specimens were monitored weekly and the environments were controlled. Then, specimens were evaluated using different mechanical testing as the Indirect Tensile (IDT) test method, compressive strength according to ASTM standards. Results of testing were documented and observed in this study for further understanding of mechanical properties of Fiber Reinforced concrete. Forensic observation of fiber distribution after the IDT tests were also performed.
