Show moreThe presence of heavy metals and other pollutants is detrimental to marine ecosystems. The queen conch, once an important fisheries species in the Florida Keys, has not sufficiently recovered after a 25-year fishery closure. Research has shown high levels of copper and zinc in the gonads and digestive glands of adult conch found in the nearshore waters. Four sites relevant to queen conch larval recruitment were tested in 2010 for the presence of copper and zinc in the water, phytoplankton, sediment, and seagrass epiphytes over seven months. Both metals were detected in all sample types and no seasonal or geographical differences were detected. Surface water concentrations from the field were used to conduct acute and chronic toxicity tests on various ages of queen conch larvae and their phytoplankton food source. When zinc concentrations (0-40 (So(Bg/L) similar to those measured in situ were used, there was no significant impact on conch larval survival although some velar lobe de velopment was impaired. However, field concentrations of copper (0-15 (So(Bg/L), which often surpassed water quality standards, negatively impacted growth, survival, and development of the larvae. Chronic exposure to copper, through the water and food, disrupted the metamorphic success of competent larvae and decreased post-metamorphosis survival. Exposure to copper at later life stages increased mortality, suggesting that heavy metals have a negative effect on larval recruitment in localized areas of the Florida Keys. Structural equation modeling revealed that copper and zinc are moving through the systems differently and are best represented by two different models.

Within the STAs, density and richness in the fall and winter were much greater in the submerged aquatic vegetation than in the mixed emergent vegetation when waterfowl were present. The STAs maintain two vegetation treatments which enhanced their biodiversity value by supporting distinct avian communities with different migratory strategies This suggests the increase in treatment wetlands could partially offset the loss of natural wetlands, but avian communities in treatment wetlands are not surrogates for natural wetlands.

The goal of this work was to explore electrosensory and visual capabilities in a more ecological context. To gain an understanding of the content of bioelectric signals, the magnitude and frequency of these stimuli were recorded from a broad survey of elasmobranch prey items... Color vision adaptations also correlated to the photic environment of each species; cownose rays inhabit turbid, green-dominated waters and had two cone visual pigments that maximize contrast of objects against the green background... Yellow stingrays were trichromatic and likely possess the ability to discriminate colors in their clear, reef and seagrass habitats, which are spectrally rich. Both species showed evidence of ultraviolet sensitivity, which may aid in predator and conspecific detection as an enhanced communication channel. Future studies should investigate the integration of sensory input and sensory involvement in intraspecific communication to gain more insight into ecological adaptations.

I used a three-part methodology to determine if color discrimination occurred. FIrst, I exposed naèive, light-adapted hatchlings to either a blue, green or yellow light. I manipulated light intensity to obtain a behavioral phototaxis threshold to each color, which provided a range of intensities we knew turtles could detect. Second, I used food to train older turtles to swim toward one light color, and then to discriminate between the rewarded light and another light color ; lights were presented at intensities equally above the phototaxis threshold. Lastly, I varied light intensity so that brightness could not be used as a discrimination cue. Six turtles completed this task and showed a clear ability to select a rewarded over a non-rewarded color, regardless of stimulus intensity. Turtles most rapidly learned to associate shorter wavelengths (blue) with food. My results clearly show loggerheads have color vision. Further investigation is required to determine how marine turtles exploit this capability.

Show moreNest predation can significantly reduce hatchling recruitment in sea turtle populations. This study uses 20 years of data from Canaveral National Seashore, Florida, which has pristine and altered beaches. Chi-square tests were used to determine if secondary predation events were related to the nest's primary depredation event. To determine if human beach use and nest predation are spatially or temporally linked, we ranked human beach use and examined predation frequency across all screened and marked nests (n=40,441). Lastly, I quantified nest predation risk spatially and temporally. I found that primary predation increases the chance of a nest suffering subsequent predation. Primary and multiple predation events occurred with greater frequency in limited use areas and with lowest frequency in moderate use areas. Predation risk decreased by an average of 29.5% from 2000-2008. Nests deposited midseason were 9.8% more likely to be predated than nests deposited early or late.

Show moreRNA damage occurring under oxidative stress has been shown to cause RNA dysfunction and must be detrimental to cells and organisms. We propose that damaged RNA can be removed by specific RNA surveillance activities. In this work, we investigated the role of polynucleotide phosphorylase (PNPase), a 3'->5' exoribonuclease, in protecting the cells against oxidative stress and eliminating oxidatively-damaged RNA. Previously, it was reported that E. coli PNPase has a higher affinity to poly(8-oxoG:A). We further confirmed that E. coli PNPase can specifically bind to an oxidized RNA with a high affinity. An E. coli strain deficient in PNPase (pnp) is hypersensitive to hydrogen peroxide (H2O2). Importantly, the level of H2O2-induced RNA damage, measured by the content of 8-hydroxyguanosine, increases significantly in the pnp mutant cells. Consistent with the notion that PNPase plays a direct role in these processes, introduction of the pnp gene encoding E. coli PNPase can restore the viability and RNA oxidation level of the pnp mutant cells in response to H2O2 treatment. Interestingly, degradosome-association is not required for PNPase to protect cell against oxidative stress. PNPase is evolutionary conserved in most of organisms of all domains of life. The human polynucleotide phosphorylase (hPNPase) localizes mainly in mitochondria and plays pleiotropic roles in cell differentiation and has been previously shown to bind 8- oxoG-RNA with a high affinity. Here we show that similar to E. coli PNPase, hPNPase plays an indispensable role in protecting HeLa cells against oxidative stress. The viability in HeLa cell and 8-oxoG levels in RNA are inversely correlated in response to H2O2- treatment. After removal of oxidative challenge, the elevated level of 8-oxoG in RNA decreases, suggesting the existence of surveillance mechanism(s) for cleaning up oxidized RNA., We have shown that hPNPase may be responsible for the surveillance of oxidized RNA in mammalian cells.Overexpresion of hPNPase reduces RNA oxidation and increases HeLa cell viability against H2O2 insult. Conversely, hPNPase knockdown decreases the viability and increases 8-oxoG level in HeLa cells exposed to H2O2. Taken together, our results suggest that RNA oxidation is a challenging problem for living organisms, and PNPase may play an important role in protecting both prokaryotic and eukaryotic cells by limiting damage to RNA under oxidative stress.

Moreover, striatal BimEL expression remained high in an R6/2 HD mouse model throughout the disease progression. Utilizing novel BimEL phospho-mutants we demonstrated the phosphorylation of Ser65 to be important for the stabilization of BimEL. We provided evidence that impaired proteasome function, increased JNK activity and reduced striatal BDNF lead to changes in the phosphorylation of BimEL, thereby promoting its stabilization specifically within the striatum of R6/2 mice. Furthermore, knocking down BimEL expression prevented mHtt-induced cell death in a HD cell culture. Taken together, these findings suggest that BimEL may contribute to the selective neurodegeneration and pathogenesis of HD.

The methodology underwent considerable changes between studies. Bass dispersed to the edges by the second sampling period, so a larger sampling area may be needed. Water depth and pH were found to be significantly different between wild and stocked bass. Hatchery bass are naèive about predators, which may have resulted in stocked bass not moving to shallower areas like wild bass. The pH was weakly correlated with depth, so differences may be partially due to the fact that as depth increases, pH may also increase.

In a large-scale experiment at the Loxahatchee Impoundment Landscape Assessment project within the Arthur R. Marshall Loxahatchee National Wildlife Refuge, I manipulated two components of prey availability, water depth and vegetation density (submerged aquatic vegetation and emergent vegetation), and quantified the response by wading birds in terms of foraging habitat selection and foraging success. Manly's standardized selection index showed that birds preferred shallow water and intermediate vegetation densities. However, the treatments had little effect on either individual capture rate or efficiency. This was a consistent pattern seen across multiple experiments. Birds selected for certain habitat features but accrued little benefit in terms of foraging success. I hypothesize that birds selected sites with shallow water and intermediate vegetation densities because they anticipated higher prey densities, but they did not experience it here because I controlled for prey density.

This study compared hydrodynamic stability between the two species during swimming to test for functional differences in body shape. Flipper movement paths, four stability measures (yaw, pitch, heave, and sideslip), and the relative positions of the centers of buoyancy and gravity were compared between species. Both centers of buoyancy and gravity lie in the anterior body; their positions relative to one another differed with species, but showed no functional consequences. Neither species demonstrated substantial yaw, sideslip, or pitch. Both experienced upward heave with the flippers' downstroke and downward heave with the upstroke; however phase relationships differed between these limb and body motions. No differences were found between the two species. Despite obvious morphological differences, loggerheads and green turtles were similarly stable during swimming, suggesting that the species use different mechanisms to achieve stability.

Selectivity analysis revealed that ibis always selected for crayfish in mixed treatments. I hypothesize that the number of prey types available in a treatment played an important role in determining how fish density and the presence of conspecifics affected ibis foraging success.

Mercury compounds are toxicants with no known biological function. High bodily concentrations of mercury can be detrimental to marine organismal health, reproduction and survival, both directly and indirectly through inducing selenium depletion. The goals of this dissertation are to evaluate several related hypotheses to explain low leatherback nest success. ... Because leatherbacks take in high volumes of prey, high tissue concentrations of mercury and selenium can result. This study provides the first evidence that chemical contaminants may explain low reproductive success in leatherback sea turtles.

This study reports on the effects of demographic changes following 2 intense hurricanes on two sympatric delphinid species: Atlantic bottlenose dolphins, Tursiops truncatus, and Atlantic spotted dolphins Stenella frontalis. Thirty percent of the bottlenose population was lost after the hurricanes, with an influx of roughly the same number of immigrants. The stable community split into two cohesive units. Preferences in association in relation to reproductive status and sex remained. Immigrants assimilated well into the population, especially males. There is a conflict of interest between resident males and females in accepting immigrants and often females find more resistance than males. Long-term analysis of spotted dolphins revealed a community structure defined by long-term site fidelity, natal philopatry of both sexes and three social clusters., Female associations were influenced by reproductive status and social familiarity within clusters. Males formed long-term alliances and shorter-term coalitions. Some movement between clusters occurred. Alliance formation crystallized in adulthood. Mating strategies and sex were the primary factors shaping social structure. Thirty-six percent of the spotted dolphin population was lost after the hurricanes with no influx of immigrants. The spotted dolphin community differed little from long-term analysis, including definitive social clusters and sex preferences. Social cohesion increased within units and across age classes. Some juveniles had associations of alliance level. Loss of individuals resulted in subtle changes in social structure. Interspecies associations reveal striking differences in association patterns between species. Group sizes and re-sightings of spotted dolphins were larger than for bottlenose dolphins, particularly for aggressive encounters., Male alliances and coalitions were prevalent for spotted dolphins, but not for bottlenose dolphins. After the hurricanes lower re-sighting and group sizes for spotted dolphin males and less aggressive behavior documented, indicates a re-structuring period of relationships between the species. This study shows that environmental variations may alter the structure of mammal societies through demographic upheaval and survival of populations may depend on their social structure and the social adaptability of the species.

In humans. Vitamin D is principally derived from sunlight induced cutaneous conversion of 7-dehydrocholesterol to vitamin D and oral supplementation. We sought to determine if established and chronic-persistent adult asthma patients from a South-Florida pulmonary patient population, with abundant sunshine availability and oral vitamin D supplementation exhibit vitamin D insufficiency/deficiency. A trend to vitamin D insufficiency was observed in approximately 65% of both adult asthma patients and apparently healthy (non-asthmatic) volunteers. . The transcription factors required for Th9 conversion, PU.1 and IRF-4, were down-regulated by vitamin D. The generation of Th9 cells was inhibited equally by vitamin D and dexamethasone when used alone, but the effect was additive when both steroids were used in combination. Our studies using non-specifically stimulated cells were extended by analyzing the effect of vitamin D on allergen specific stimulation. The response of CD4+ T cells obtained from the blood of house dust mite positive asthmatics was studied. House dust mite allergen elicited a classical Th2 phenotype response (IL-4, IL-5, IL-9, and IL-13 cytokine profile) and vitamin D effectively inhibited those key Th2 cytokines. We conclude that vitamin D appears to be of significant clinical benefit in our cohort of patients, i.e., established chronic adult human asthma, by down-regulating key immune cells including Th9, Th17, and Th2 involved in this disorder.

Orientation was unchanged when the turtles bore magnets that negated the use of magnetic cues. These results are consistent with the hypothesis that the turtles used the sun for orientation.

This study aimed to reveal the symbiosis between AM fungi and sawgrass and cattail and identify the phosphate transporters, especially AM-specific phosphate transporters in these two species. AM colonization was only found in sawgrass roots, not cattail, at low phosphate concentrations in lab and field samples by trypan blue staining. AM fungi could increase sawgrass growth and had little effect on cattail growth. Four phosphate transporters were identified in sawgrass. CjPT1, CjPT2 and CjPT3 were expressed in roots and shoots independent of AM fungi and phosphate availability, while CjPT4 appeared to be an AM regulated phosphate transporter gene and its expression was induced by AM fungi.

Results of this research project suggest that the effect of sulindac and oxidative stress not only involves mitochondrial ROS production, but also aspects of the preconditioning response. In normal cells this leads to survival by a preconditioning pathway, likely involving PKCε. . However, cancer cells react by initiating a pathway leading to apoptosis involving PKCδ.

Show moreThere are four distinct subpopulations of loggerhead sea turtles (Caretta caretta) in Florida as determined behaviorally by geographic fidelity, and genetically by mitochondrial haplotypes. The South Florida subpopulation consists of females nesting on the southeastern and southwestern coasts of Florida and their offspring. Previous research shows that west coast hatchlings exhibit higher levels of nocturnal swimming during the postfrenzy period than east coast hatchlings. This study attempted to determine how these differences in migratory behavior develop. A reciprocal translocation experiment was conducted to distinguish between environmental and genetic factors. No consistent differences in hatchling swimming behavior were seen based on geography. Movement of nests resulted in lower levels of nocturnal swimming behavior in hatchlings compared to hatchlings that emerged from natural nests, suggesting that the relocation of nests may not provide a natural incubation environment for developing hatchlings.

Show moreBackground: Light-adaptation is a multifaceted process in the retina that helps adjust the visual system to changing illumination levels. Many studies are focused on the photochemical mechanism of light-adaptation. Neural network adaptation mechanisms at the photoreceptor synapse are largely unknown. We find that large, spontaneous Excitatory Amino Acid Transporter (EAATs) activity in cone terminals may contribute to cone synaptic adaptation, specifically with respect to how these signals change in differing conditions of light. EAATs in neurons quickly transport glutamate from the synaptic cleft, and also elicit large thermodynamically uncoupled Cl- currents when activated. We recorded synaptic EAAT currents from cones to study glutamate-uptake events elicited by glutamate release from the local cone, and from adjacent photoreceptors. We find that cones are synaptically connected via EAATs in dark ; this synaptic connection is diminished in light-adapted cones. Methods: Whole-cell patch-clamp was performed on dark- and transiently light-adapted tiger salamander cones. Endogenous EAAT currents were recorded in cones with a short depolarization to -10mV/2ms, while spontaneous transporter currents from network cones were observed while a local cone holding at -70mV constantly. DHKA, a specific transporter inhibitor, was used to identify EAAT2 currents in the cone terminals, while TBOA identified other EAAT subtypes. GABAergic and glycinergic network inputs were always blocked with picrotoxin and strychnine. Results: Spontaneous EAAT currents were observed in cones held constantly at -70mV in dark, indicating that the cones received glutamate inputs from adjacent photoreceptors. These spontaneous EAAT currents disappeared in presence of a strong light, possibly because the light suppressed glutamate releases from the adjacent photoreceptors. The spontaneous EAAT currents were blocked with TBOA, but not DHKA, an inhibitor for EAAT2 subtype, suggesting that a, non-EAAT 2 subtype may reside in a basal or perisynaptic area of cones, with a specialized ability to bind exocytosed glutamate from adjacent cones in dark. Furthermore, these results could be artificially replicated by dual-electrode recordings from two adjacent cones. When glutamate release was elicited from one cone, the TBOA-sensitive EAAT currents were observed from the other cone. Conclusions: Cones appear to act like a meshwork, synaptically connected via glutamate transporters. Light attenuates glutamate release and diminishes the cone-cone synaptic connections. This process may act as an important network mechanism for cone light adaptation.

Activins are involved in regulating steroid hormone release in vertebrates. In invertebrates, they most likely function to keep ecdysone levels low to allow the larvae more time to achieve critical weight in order to initiate the metamorphic process. TGF-B(Transforming Growth Factor Beta) is a family of cytokine growth factors. We find that two members of the TGF-B signaling pathway Drosophila Activin (dACT) and Activin-like ligand Dawdle (DAW) signal through the type I receptor Baboon (BABO) and the type II receptor PUNT to primarily activate the transcription factor dSMAD2 and MAD to a lesser extent. One transcription factor brinker (brk) appears to be central to dACT signaling., In wings dACT signaling is necessary to promote growth however, dACT is not expressed in wings suggesting that dACT is provided through the endocrine system. One possible target tissue of dACT signaling is the ring gland (RG), which synthesizes and secretes the steroid hormone ecdysone (E). Consistent with this idea, using the UAS/GAL-4 system, we find that over-expression of the TGF-B ligand dACT with the neuroendocrine driver 386Y-GAL4 results in an increase in the size of flies. Surprisingly, when we increase the dose with two copies of dACT, it decreases the size of flies also indicating non-autononomous effects. We find that overexpression of the activated form of the dACT type I receptor Baboon (BABO) or brk with the ring gland specific driver phm-GAL4 results in developmental arrest of larvae that stay small and never pupate. The developmental arrest can be overcome by feeding larvae E, suggesting that dACT represses E through brk. These results suggest a model where dACT signaling activates brk which inhibits E. We picked three cytochrome P450 enzymes: phantom (PHM), disembodied (DIS) and spookier (SPKR)., PHM is not regulated by any component in the dACT signaling pathway however, we find DIS and SPKR are down-regulated through brk. MAD and dSmad2 bind to a Smad binding site and MAD out-competes dSMAD2. We find no evidence that Drosophila insulin-like peptides (DILPS)/PI3- Kinase or Ras signal through the dActivin signaling pathway.
