Show moreTranscription factors are macromolecules that are involved in transcriptional regulation by interacting with specific DNA regions, and they can cause activation or silencing of their target genes. Gene regulation by transcriptional control explains different biological processes such as development, function, and disease. Even though transcriptional control has been of great interest for molecular biology, much still remains unknown. This study was designed to generate the most current list of human transcription factor genes. Unique entries of transcription factor genes were collected and entered into Microsoft Office 2007 Access Database along with information about each gene. Microsoft Office 2007 Access tools were used to analyze and group collected entries according to different properties such as activator or repressor record, or presence of certain protein domains. Furthermore, protein sequence alignments of members of different groups were performed, and phylogenetic trees were used to analyze relationship between different members of each group. This work contributes to the existing knowledge of transcriptional regulation in humans.

Show moreActivity patterns of Iguana iguana from two locations within Hugh Taylor Birch State Park, Fort Lauderdale, Florida were documented and examined. Between May 1, 2006 to April 20, 2007, I. iguana were observed on a routine basis and activities were documented as one of six major activity categories (basking, locomotion, foraging, resting, visual signaling, and other). Data was analyzed to determine differences between activity patterns of I. iguana relative to sites, seasons, and size category within the park. Iguana iguana spent more time basking at Site 1 than Site 2. Size 4 animals which consisted of dominant adult males, spent more time basking than other males and adult females. Size 4 animals also spent less time foraging than hatchlings, juveniles, and other adults. These results complement the existing research on behavior of I. iguana and may be useful in determining invasive control efforts of I. iguana in Florida.

Despite their reputation for being particularly olfactory driven, little is known about how this sense functions in elasmobranch fishes. The goal of this dissertation was to examine the morphology and physiology of elasmobranchs to compare their olfactory system with teleost fishes and more derived vertebrates. To test the hypotheses that elasmobranchs possess greater olfactory sensitivities than teleosts and that lamellar surface area is correlated to sensitivity, I compared the surface area of the olfactory lamellae and the olfactory sensitivities of five phylogenetically diverse elasmobranch species. The olfactory thresholds reported here (10-9 to 10-6 M) were comparable to those previously reported for teleosts and did not correlate with lamellar surface area. Since aquatic species are subject to similar environmental amino acid levels, they appear to have converged upon similar amino acid sensitivities. To test the hypothesis that elasmobranchs are able to detect bile salt odorants despite lacking ciliated olfactory receptor neurons (ORNs), the type of ORN that mediates bile salt detection in the teleosts, I quantified the olfactory specificity and sensitivity of two elasmobranch species to four, teleost-produced C24 bile salts. Both species responded to all four bile salts, but demonstrated smaller relative responses and less sensitivity compared to teleosts and agnathans. This may indicate that elasmobranchs don't rely on bile salts to detect teleost prey. Also, the olfactory system of elasmobranchs contains molecular olfactory receptors for bile salts independent of those that detect amino acids, similar to teleosts., In some elasmobranch species, each olfactory bulb (OB) is physically partitioned into two hemi-bulbs; however, the functional significance of this morphology is not fully understood. The organization of the OBs in three species with varying OB morphologies was examined to test the hypothesis that the elasmobranch OB is somatotopically arranged. Glomeruli in the OB received projections from ORNs in 3-4 olfactory lamellae situated immediately anterior. These results indicate a somatotopic arrangement of the elasmobranch OB, which may be unique among vertebrate olfactory systems and potentially led to the hemi-OB morphology.

The cellular mechanisms underlying the anatomical plasticity of the adult olfactory system are not well understood, but neurotrophins, long recognized as crucial to the early development of neuronal circuits, are attractive candidate molecules in this capacity. Brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF), two members of the neurotrophin family, have received attention in terms of their anatomical distribution and physiological functions in the olfactory system.

The potential efficacy and mechanism of topotecan/genistein-induced cell death was investigated... Results: The overall data indicated that i) both genistein and topotecan induce cellular death in LNCaP cells, ii) topotecan-genistein combination was significantly more efficacious in reducing LNCaP cell viabiligy compared to either genistein or topotecan alone, iii) in all cases, cell death was primarily through apoptosis, via the activation of the intrinsic pathway, iv) ROS levels were increased and VEGF expression was diminished significantly with the topotecan-genistein combination treatment, v) genetic analysis of topotecan-genistein treatment groups showed changes in genetic expression levels in pathway specific apoptotic genes.... Conclusion: Treatments involving topotecan-genistein combination may prove to be an attractive alternative phytotherapy of adjuvant therapy for prostate cancer.

The percentage of apoptosis was higher in combination treatments than single treatments with either phytochemical, although the difference was not statistically significant. The combination of genistein and anthocyanin extract peaked in efficacy at 48 hours of treatment, to exhibit significantly greater (P<. O5) dose- and time-dependent cell cytotoxicity than single treatments. This study reveals potential chemopreventive implications for the complementary effects of genistein and anthocyanin extract.

Show moreThe tunicate Styela plicata (Ascidiacea) was investigated for its potential use in bioremediation of bacteria and microalgae blooms from estuarine waters. Salinity tolerances, filtration rate, substrate selection, and effects on digested bacteria and ichthyofaunal communities were examined. If acclimated slowly, Styela can be placed in salinities as low as 24 parts per thousand (ppt) before increased fatalities result. An immediate decrease in salinity from 32 ppt to 20 ppt was not detrimental at short term exposure (four days). One average-sized (~40g) Styela, exposed to 105 and 106 bacteria or microalgae mL-1, can filter as much as 4.7 L hr-1 and 3.3 L hr-1, respectively. Individual tunicate filtration rates varied from hour to hour, independent of organism size, indicating that filtration rates for this species would be better reported on a population basis rather than on an individual weight or size basis. Bacterial viability in tunicate feces was assessed by ATP analysis. Bacteria were found to be non-viable after passage through the tunicate digestive tract. Filtration of eggs or larvae of recreationally or commercially important fish is of concern. The smallest eggs (~0.6mm) reported in the Indian River Lagoon belong to Cynoscion nebulosus (spotted seatrout) and Bairdiella chrysoura (silver perch). Over the course of two hours, 72.5% of 0.5 mm glass beads (simulated fish eggs) were initially retained, but many were later expelled by the tunicates. For 1.0 mm beads, 49.4% were initially retained and for 2.0 mm beads, 43.5% were initially retained. Neither the size of the oral siphon nor the size of the overall animal was correlated to bead retention. Substrate selection was investigated using the tunicate Phallusia nigra., Newly hatched larvae preferred settling on wood (53.7%), as compared to polyethylene (30.9%), high-density polyethylene (13.0%), and polyvinyl chloride (2.4%). Overall, results of this study indicate S. plicata is very effective at removing bacteria and microalgae from estuarine waters. However, further testing is warranted regarding the unwanted removal of fish eggs and larvae before utilization of S. plicata for bioremedial purposes.

The STAs contained a more abundant, rich and distinct avian community compared to reference land types. The STAs were dominated by wintering waterfowl, and therefore community patterns fluctuated more seasonally other land types. Within the STAs, density and richness in the fall and winter were much greater in the submerged aquatic vegetation than in the mixed emergent vegetation when waterfowl were present. The STAs maintain two vegetation treatments which enhanced their biodiversity value by supporting distinct avian communities with different migratory strategies This suggests the increase in treatment wetlands could partially offset the loss of natural wetlands, but avian communities in treatment wetlands are not surrogates for natural wetlands.

Although no treatment effect was found, catalase activity was enhanced in T. testudinum leaves and H. wrightii roots relative to other tissues, while S. filiforme showed no location-specific catalase activity. These results indicate that quantum efficiency is a more sensitive indicator than protein and catalase activity to hypoxia and sulfide stress in seagrasses.

A majority of transcriptional repressors are zinc finger proteins (ZFPs) which bind to specific DNA sequences. The Snail/Gfi (SNAG) domain family, with members such as Slug, Smuc, Snail, and Scratch, are transcriptional repressors shown to play a role in various diseases such as cancer. The SNAG transcription factors contain a conserved SNAG repression domain and DNA binding domain zinc fingers. The specific DNA sequences to which each SNAG-ZFP binds, as well as a general consensus -TGCACCTGTCCGA, have been determined. Also, putative protein-protein interactions in which the Slug domain participates has been identified via binding assays. All these results contribute to better understanding of SNAG-ZFP functions.

Geographic genetic variation is suggested by the presence of population specific alleles. The results showed considerable divergence of groups of Bald Eagle alleles when compared to alleles from other birds of prey. Particular codons within the exon II show signs of balancing selection driving the evolution of the MHC II B. Transcription data showed statistically significant differential expression of alleles. This can be interpreted as meaning a particular locus is being preferentially expressed in blood. The analysis of the polymorphism of this adaptive marker may aid managers of wildlife during this age of global climate change and the biodiversity crisis.

Show moreBrain glutamic acid decarboxylase 65 (GAD65) catalyzes the rate-limiting step in the biosynthesis of the major inhibitory neurotransmitter-amino butyric acid (GABA) from the substrate L-glutamic acid. Severe lapse in GABA neurotransmission is one of the etiologies documented in the manifestation of certain neurodegenerative diseases such as epilepsy, Parkinson's disease, Huntington's disease etc. Because GAD65 synthesizes GABA, any modulation of GAD65, therefore, has direct implications on the quanta of GABA released at the synapse. Hence, the major objective of this study was to focus on the regulation of GAD65, with special emphasis on investigating the proteolytic cleavage of fGAD65. Previously, we have shown in vitro that GAD65 was cleaved to form its truncated form (tGAD65), which was more active than the full length form (fGAD65). The enzyme responsible for cleavage was later identified as calpain. Calpain is known to cleave its substrates either under a transient physiologica l stimulus or upon a sustained pathological insult. However, the precise role of calpain cleavage of fGAD65 is poorly understood. In this study, we examined the cleavage of fGAD65 under a range of conditions encompassing both physiological and pathological aspects, including rats under ischemia/reperfusion insult, rat brain synaptosomes or primary neuronal cultures subjected to excitotoxic stimulation with KCl. It was observed that the formation of tGAD65 progressively increased with increasing stimulus concentration. More importantly, cleavage of synaptic vesicle (SV) - associated fGAD65 by calpain was demonstrated, and the resulting tGAD65 harboring the active site of the enzyme was detached from the SVs. Vesicular uptake of the newly synthesized GABA into the SVs was found to be reduced in calpain treated SVs. Furthermore, we also observed that the levels of tGAD65 in the focal cerebral ischemic rat brain tissue increased corresponding to the elevation of local glutamate indica, d by in vivo micro dialysis. Based on these observations, we conclude that calpain cleavage of fGAD65 occurs under pathological conditions.

Show moreMacroalgal blooms are responses to nutrient enrichment in shallow seagrass ecosystems like the Indian River Lagoon (IRL), Florida. Little is known about nitrogen (N) and phosphorus (P) limitation or the importance of morphological/physiological characteristics of bloom-forming macroalgae (Ulva lactuca, Hypnea musciformis, and Gracilaria tikvahiae) in the IRL. We hypothesized: 1) all species would proliferate in nutrient-rich Titusville, 2) opportunistic U. lactuca would dominate, 3) Rapid Light Curves (RLCs) would assess nutrient status, and 4) nutrient concentrations would regulate growth more than N:P ratios. Field studies showed rapid biomass doubling times of 2 days (U. lactuca; November 2012) in urbanized Titusville. RLCs in a guano-enriched island off Big Pine Key (BPK) and Titusville (Ulva spp.) were similar due to P-saturation. Laboratory studies showed three-fold higher RLCs and two-fold faster growth at high nutrient concentrations of N and P. Reductions of both N and P will be required to moderate future blooms.

This study compared hydrodynamic stability between the two species during swimming to test for functional differences in body shape. Flipper movement paths, four stability measures (yaw, pitch, heave, and sideslip), and the relative positions of the centers of buoyancy and gravity were compared between species. Both centers of buoyancy and gravity lie in the anterior body; their positions relative to one another differed with species, but showed no functional consequences. Neither species demonstrated substantial yaw, sideslip, or pitch. Both experienced upward heave with the flippers' downstroke and downward heave with the upstroke; however phase relationships differed between these limb and body motions. No differences were found between the two species. Despite obvious morphological differences, loggerheads and green turtles were similarly stable during swimming, suggesting that the species use different mechanisms to achieve stability.

Show moreTranscriptional regulation by the family of SNAG (Snail/Gfi-1) zinc fingers has been shown to play a role in various developmental states and diseases. These transcriptional repressors have function in both DNA- and protein-binding, allowing for multiple interactions by a single family member. This work aims to characterize the SNAG members Slug, Smuc, Snail, Scratch, Gfi-1, Gfi-1B, and IA-1 in terms of both DNA-protein and protein-protein interactions. The specific DNA sequences to which the zinc finger regions bind were determined for each member, and a general consensus of TGCACCTGTCCGA, was developed for four of the members. Via these studies, we also reveal thebinding affinities of E-box (CANNTG) sequences to the members, since this core is found for multiple members' binding sites. Additionally, protein-protein interactions of SNAG members to other biological molecules were investigated. The Slug domain and Scratch domain have unknown function, yet through yeast two-hybrid screening, we were able to determine protein interaction partners for them as well as for other full length SNAG members. These protein-interacting partners have suggested function as corepressors during transcriptional repression. The comprehensive information determined from these studies allow for a better understanding of the functional relationship between SNAG-ZFPs and other genes. The collected data not only creates a new profile for each member investigated, but it also allows for further studies to be initiated from the results.

I have constructed glutathione S-transferase (GST)-ZAD domain chimeric proteins for use in pull down protein binding assays,and GST-Zinc finger (ZnF) array domain chimera for electrophoretic mobility shift assays (EMSA). Protein binding assays indicated two putative conserved interactors, similar to the analogous KRAB system in mammals. ... Competitive bindings were carried out to show a specificity of binding conferred by the identified conserved positions. While the consensus binding sites show relatively few similarities, the predicted target genes identified by the consensus binding sites show significant overlap. The nature of this overlap conforms to the known characteristics of the ZAD family but points to a more positive selection to maintain conservation of function.

I examined the effect of sunfish predators on two species of south Florida crayfish to establish the mechanism that limits one species, Procambarus alleni, to short-hydroperiod environments. The crayfish assemblage response to a gradient of sunfish predators and the effect of predation on P. fallax alone was examined. I also examined the effects of sunfish on crayfish growth and quantified activity levels and risky behaviors of both crayfish species. P. alleni dominated at low sunfish densities but dominance shifted with increasing sunfish density. P. alleni was more active and likely to initiate risky behaviors, suggesting that sunfish predators remove the more active P. alleni, reducing their numbers disproportionally to those of P. fallax and allowing P. fallax to dominate crayfish assemblages in long-hydroperiod wetlands.

Show moreThe central premise of this dissertation is that the small heat shock protein (sHSP), (Sa(BB-crystallin is essential for lens and retinal pigmented epithelial (RPE) cell function and oxidative stress defense. To date, the mechanism by which it confers protection is not known. We hypothesize that these functions could occur through its ability to protect mitochondrial function in lens and RPE cells. To test this hypothesis, we examined the expression of (Sa(BB-crystallin/sHSP in lens and RPE cells, we observed its localization in the cells, we examined translocation to the mitochondria in these cells upon oxidative stress treatment, we determined its ability to form complexes with and protect cytochrome c (cyt c) against damage, and we observed its ability to preserve mitochondrial function under oxidative stress conditions in lens and RPE cells. In addition to these studies, we examined the effect of mutations of (Sa(BB-crystallin/sHSP on its cellular localization and translocation patterns under oxidative stress, its in vivo and in vitro chaperone activity, and its ability to protect cyt c against oxidation. Our data demonstrated that (Sa(BB-crystallin/sHSP is expressed at high levels in the mitochondria of lens and RPE cells and specifically translocates to the mitochondria under oxidative stress conditions. We demonstrate that (Sa(BB-crystallin/sHSP complexes with cyt c and protects it against oxidative inactivation. Finally, we demonstrate that (Sa(BB-crystallin/sHSP directly protects mitochondria against oxidative inactivation in lens and RPE cells. Since oxidative stress is a key component of lens cataract formation and age-related macular degeneration (AMD), these data provide a new paradigm for understanding the etiology of these diseases.

For this, we paired nanoinjection of compounds with electrophysiological recordings from the Giant Fiber System of Drosophila melanogaster, which mediates the escape response of the fly.

Identifying primary prey types of nesting wading birds is important for the hydrologic restoration of wetlands. I collected nestling boluses during the 2008 and 2009 nesting seasons from three species of wading birds that nest in the northern Everglades: White Ibis, Tricolored Herons and Snowy Egrets. White Ibis bolus composition was dominated by crayfish in both years, but exhibited some variation with landscape water depth in 2009; fish use was greatest when the wetland landscape was relatively dry. In contrast, the prey of Tricolored Herons and Snowy Egrets were primarily fish and their respective diets did not differ from one another in either fish species composition or size structure.
