By examining the symbiotic zooxanthellae, or Symbiodinium, that reside in corals, we can gain insight to coral physiology impacted by local stressors. Two scleractinian corals, Montastraea cavernosa and Pseudodiploria clivosa were sampled over 1.5 years, including both wet and dry seasons. Zooxanthellae were isolated and quantitatively characterized using standard measurements and molecular techniques. Both coral species varied in zooxanthellae biomass, where Pseudodiploria clivosa had Higher cell densities and chlorophyll concentrations than Montastraea cavernosa. Over time, these parameters varied, but were not significantly altered by fresh water discharge events. Symbiodinium diversity and abundance were identified by ITS2 region amplification and next-generation sequencing .Novel associations between Symbiodinium and each coral explained the observed physiological differences. The symbioses remained stable throughout and could indicate local adaptation for St. Lucie Reef corals.

The secondary metabolites of B. asbestinum fall into four classes : asbestinins, briarellins, briareolate esters, and briaranes. Briareolate esters have been shown to possess biological activity and were previously only reported from Tobago. Our group recently isolated briareolate esters from a specimen collected off the coast of Boca Raton, Florida. To determine whether location has an impact on the chemistry produced by the organism, a method to discern between chemotypes was sought. Several techniques including thin layer chromatography (TLC), high performance liquid chromatography (HPLC), nuclear magnetic resonance (NMR), and sclerite analysis were employed, with NMR being the most successful method. By utilizing both 1H and COSY NMR experiments, it is possible to differentiate between the chemotypes of B. asbestinum. Application of this method allowed analysis of chemical variability with respect to location.

Show moreClimate change has intensified thermal anomalies in coral reef ecosystems, contributing to coral bleaching and decline. As corals die, reef fragmentation increases, and species interactions in the benthos change. However, it is unclear which competitive interactions may prevail and structure future reef ecosystems. The aim of this thesis was to evaluate the effect of thermal anomalies on coral reef benthic competitive interactions. Photoquadrats in southeast Florida reef sites were assessed over 15-years to generate interaction metrics and determine effects on hard coral survivorship. A state-transition model was created to predict the resultant community across 100 years with thermal scenarios concordant with the IPCC RCP 4.5 and 8.5. Interaction doubled across three thermal anomaly events and ended up mainly composed of pairwise examples between Dictyota, Halimeda, Niphates erecta, and Erythropodium caribaeorum. Century projections confirm that soft coral and sponge interactions will increase through thermal anomalies. The survival of hard corals was more successful when colonies were in permanent or intermittent interactions than when colonies were solitary (indirect interactions). Living hard corals were mostly found interacting with the macroalgae, Dictyota, and sponge Aplysina cauliformis, while corals that died were mainly in interactions with the soft coral E. caribaeorum, and sponges N. erecta, C. delitrix, D. anchorata, and Ircinia campana. Future reefs will be composed of more interactions between soft corals and sponges as thermal anomalies intensify, which will result in a patchier and flatter community.

Show moreThe overall goal of this research was to isolate key genes involved in the diterpene biosynthesis from Euniceafusca and Erythropodium caribaeorum using molecular biology techniques. The initial goal was to use fuscol induced cell cultures of Symbiodinium sp. isolated from E. fusca and to develop an approach based on differential display of mRNA-reverse transcription-PeR. Together with inverse PCR, these techniques ultimately provided a full-length farnesyl diphosphate synthase sequence. Functional expression of this enzyme was demonstrated with the addition of appropriate substrates and confirmed by chromatography. From this data, degenerate primer based PCR was used to isolate putative geranylgeranyl diphosphate biosynthetic genes from E. caribaeorum. Both chemical and genetic examinations of Pseudopterogorgia elisabethae eggs and their associated Symbiodinium sp. were employed to identify the biosynthetic origin of their diterpenes. Terpene content and biosynthetic capabilities of azooxanthellae eggs demonstrated the presence of pseudopterosins but also indicated that the eggs were not capable of producing these compounds. Likewise, no correlation could be observed for the phylogenetic relationships inferred for the Symbiodinium sp., with that of the terpene chemistry present in P. elisabethae. This finding leads us to speculate about an additional source of terpene production within this coral. Based on these and other recent findings suggesting symbiotic bacteria as the source of secondary metabolites from marine invertebrates, bacterial assemblages from E. caribaeorum were examined. This study revealed considerable phylogenetic bacterial diversity within this coral and the identification of several bacteria known to produce terpenes in other organisms.

Show moreThe impact of grazers on the primary production of marine ecosystems has largely been explored in tropical environments. A number of studies support theories on the functional importance of grazers in the community structure of coral reefs. However, large-bodied grazers, like juvenile green turtles, co-occur with herbivorous fishes in subtropical and tropical regions throughout the world and we know little about their combined impact on macroalgal communities and whether they compete for macroalgal resources. My dissertation research was composed of four studies that were conducted simultaneously to further our understanding of plant/herbivore interactions in marine ecosystems. Studies were conducted at the Trident Basin, a non-public military facility within the Port Canaveral Inlet at Cape Canaveral, Florida, USA. The macroalgal study (Chapter 1), determined the spatial and temporal distribution of the macroalgal community. The foraging habits of juvenile green turtles were compared with the macroalgal abundance within the Basin and over time (Chapter 2). Selection ‘for’ specific macroalgal species (based on their availability in the macroalgae study) was used to determine the level of overlap and/or partitioning of resources among herbivorous fishes and juvenile green turtles (Chapter 3). The final empirical study (Chapter 4) measured the impact on thallus height, diameter and/or branching of macroalgae as well as the macroalgal community composition from caging experiments that excluded herbivorous fishes and juvenile green turtles. The algal community was predominantly composed of nine red and green macroalgal species that were persistent year-round. Grazer-resistant macroalgae were rarely observed. Green turtles foraged on many of these same macroalgae but also opportunistically foraged on flotsam, including anthropogenic debris (e.g., plastic). The gut content of the major herbivorous fishes in the community (Abudefduf saxatilis, Archosargus probatocephalus, Diplodus holbrooki, and Lagodon rhomboides) foraged as omnivores depending on where they were captured within the Basin area or their size. All herbivores showed selection for less abundant green algae (i.e., Ulva spp.). Results of the exclusion of juvenile green turtles and large herbivorous fishes in caging experiments suggest that grazing by these large-bodied herbivores had no impact on the composition of the macroalgal community and little impact on the morphological structure of the macroalgal species that were examined. Collectively these four studies contribute to a better understanding of how multiple grazers have evolved to forage in macroalgal communities without detrimental effects on their food resources.

Stationary fish census coupled with benthic transect videos were conducted quarterly between April 2005-July 2007. A significant correlation between Lyngbya sp. blooms and alterations in fish assemblages was observed. Lyngbya had a negative interaction with fish species and abundance; additionally, HAB sample periods were significantly different from low algal abundance periods. Blooms of Dictyota sp. had no measurable relationship with the fish assemblage indicating HABs may have variable impacts on fishes depending on family, species and chemistry of the algae.

Digital analysis of the high-resolution (4 m) IKONOS imagery employed ESRI's ArcMap to manually digitize 412 mapping units at a scale of 1:6,000 differentiated by spectral reflectance, color tones, and textures of seafloor topologies. The context of each morphodynamic zone is characterized by the content and areal distribution (in km2) of geomorphic forms and biological covers. Over 58% of the mapping area is occupied by sediment flats, and seagrasses are colonized in almost 80% of the topologies.

Recently, two new briarane diterpenoids, briareolate esters J and K, together with two known briareolate esters have been isolated from a specimen of Briareum asbestinum collected off the coast of Boca Raton, Florida. The method used was a 96-well plate real-time cell electronic sensing (RT-CES) system to discover compounds that impact human embryonic stem cell growth. The compounds were isolated using reversed phase polystyrene divinylbenzene chromatographic support HP20ss followed by normal phased HPLC using a luna silica column. The structures of the compounds were established though the interpretation of spectroscopic data. Activity testing was conducted against hESCs (BG02) with briareolate ester J showing no inhibition activity and briareolate ester K showing mild activity with an EC50 value of 25 (So(BM. These results confirm that the exact confirmation and existence of the (E,Z)-dienone is related to the activity that was observed with the previously isolated briareolate esters L and M.

One major factor contributing to these changes is the movement of sand. This is influenced by location-based, human-induced, and natural factors. These shifting sands cover or uncover reef structure. Images of the Florida Reef Tract in Palm Beach County were analyzed by the Department of Geosciences at Florida Atlantic University, using a partially automated method of mapping. There are notable changes in reef structure throughout the years 2004-2006, in which many major storm events occurred in the region. A time series analysis was conducted throughout these years. Losses and gains of reef structure were quantified and compared throughout the county as a whole, in beach renourishment project areas, and inlet intervals. Trends suggest that the major storms of 2004-2006 may have had effects on the reef tract.

Nematocysts were identified by type and size to examine the potential plasticity associated with nematocyst production. While no significant differences were found in defensive nematocyst concentration between shrimp predation treatments versus controls, there was a significant difference in small-sized nematocyst in anemones damaged with forceps. The proportions of the different types of nematocysts between treatment types were also found to be different suggesting that nematocyst production in A. pallida is a plastic trait.

These results suggest that high APA observed in carbonate-rich waters of Dade County and low APA in Broward County resulted from high nutrient inputs associated with anthropogenic nutrient pollution.

Under rapid climate change, a clear understanding of how reefs and their benthic organisms respond is still lacking and needs to be investigated. Using in situ imagery, a sponge cell model, and long-term benthic biota surveys, the effects of climate change on reef dynamics were explored in this dissertation project. Results from the in situ imagery found that differences in spectral signatures are found between functional groups (i.e., corals, sponges, and algae) and different species from substrate. Results based on a sponge cell model and transcriptomics data have found a resilience of these sponges to the predicted thermal extremes. Results from benthic biota surveys suggested that depth and light attenuation have the largest influence on the predicted distribution of corals, sponges, and algae at Pulley Ridge. Climate change has been impacting reef benthic biota starting at the organismal scale up to the reef scale. This research demonstrates the importance of monitoring reefs at a finer scale and determining the thresholds and limits of benthic biota to projected thermal extremes to better inform resource managers to preserve these irreplaceable ecosystems.

Here I show that anemones hosting symbiotic alpheid shrimp are extremely less likely to sustain damage by predators than solitary anemones. The role of protector may be reciprocally provided by anemone and decapod associates.

The deep-sea fish family Melamphaidae (Stephanoberyciformes) is one such example of an understudied group of fishes. Samples from the MAR-ECO (www.mar-eco.no) project represent one of the largest melamphaid collections, providing an ideal opportunity to gain information on this important, but understudied, family of fishes. The key to the family presented here is the first updated, comprehensive key since those produced by Ebeling and Weed (1963) and Keene (1987). Samples from the 2004 MAR-ECO cruise and the U.S. National Museum of Natural History provided an opportunity to review two possible new species, the Scopelogadus mizolepis subspecies, and a Poromitra crassiceps species complex. Results show that Scopeloberyx americanus and Melamphaes indicoides are new species, while the two subspecies of Scopelogadus mizolepis are most likely o nly one species and the Poromitra crassiceps complex is actually several different species of Poromitra. Data collected from the MAR-ECO cruise provided an opportunity to study the distribution, reproductive characteristics and trophic ecology of the family Melamphaidae along the Mid-Atlantic Ridge (MAR). Cluster analysis showed that there are five distinct groups of melamphaid fishes along the MAR. This analysis also supported the initial observation that the melamphaid assemblage changes between the northern and southern edges of an anti-cyclonic anomaly that could be indicative of a warm-core ring. Analysis of the reproductive characteristics of the melamphaid assemblage revealed that many of the female fishes have a high gonadosomatic index (GSI) consistent with values found for other species of deep-sea fishes during their spawning seasons., This may indicate that melamphaids use this ridge as a spawning ground. Diets of the melamphaid fishes were composed primarily of ostracods, a mphipods, copepods and euphausiids. Scopelogadus was the only genus shown to have a high percent of gelatinous prey in their digestive system, while Melamphaes had the highest concentration of chaetognaths. This work presents data on the ecology and taxonomy of the family Melamphaidae and provides a strong base for any future work on this biomass-dominant family of fishes.

Elevated pCO2 under natural reef diurnal CO2 cycles did not significantly affect CCA percent cover, calcification rates or survival of adult CCA lobes. No significant community pCO2 effects were observed, rather light controlled dominance. The percent cover of microalgae increased in highlight, while CCA increased in the shade. My results indicate that algal response to irradiance is a more significant driver of reef benthic algal change than pCO2 levels predicted for 2100; however, this conclusion should be corroborated in longer-term and in field experiments.
