Water, Hoagland solution, glucose solutions, kinetin and benzimidazole were tested for their ability to delay senescence of detached Eichhornia crassipes leaves. Water and Hoagland solution failed to sustain chlorophyll past 9 days. Glucose solutions supported contaminating microbial growth and were judged unsatisfactory. Kinetin and benzimidazole solutions maintained a "normal appearance" in detached leaves beyond three weeks culture. Determination of the conversion of protein to amino acids...

Water and Hoagland solution failed to sustain chlorophyll past 9 days. Glucose solutions supported contaminating microbial growth and were judged unsatisfactory. Kinetin and benzimidazole solutions maintained a "normal appearance" in detached leaves beyond three weeks culture. Determination of the conversion of protein to amino acids and amines as well as carbohydrate levels in detached leaf culture indicated the optimal culture media were 100 ppm benzimidazole and benzimidazole-kinetin mixture, 50 ppm each. The effective length of culture was three weeks. The utility of this leaf culture system as an alternative to intact host inoculation with pathogenic microorganisms is demonstrated by the isolation and subculture of a pathogenic Alternaria in E. crassipes leaves supported by 50 ppm benzimidazole solution.
