Show moreIn this thesis the transition region between two modes of behavior is explored using a novel technique, delayed feedback, and a variety of dynamical systems measures. In a previous study, Engstrom, Kelso, and Holroyd (to appear) established the existence of a transition between anticipatory and reactive behavior in a sensorimotor coordination task as a control parameter (frequency) was varied. Here, in order to explore the hypothesis that the behavioral dynamics during this transition are intermittent in character, subjects were asked to synchronize with a metronome that was actually a delayed copy of their own response pattern. The use of delayed feedback was expected to destabilize the behavioral dynamics enough to allow the observation of hypothesized intermittent phenomena. Use of delayed feedback was shown to destabilize synchronization, resulting in the emergence of a new behavioral pattern in the transition region that exhibited complex "bursting" dynamics. Analysis revealed that this bursting behavior displays many of the characteristics common to intermittency, which supports the idea that the anticipation-reaction transition is the result of a neurobehavioral dynamical system losing stability. Living in the vicinity of instabilities may be an important mechanism for biological organisms to maintain both flexibility and stability.

Show moreModeling a biological systems, is a cyclic process which involves constructing a model from current theory and beliefs and then validating that model against the data. If the data does not match, qualitatively or quantitatively then there may be a problem with either our beliefs or the current theory. At the same time directly finding a model from the existing data would make generalizing results difficult. A considerable difficultly in this process is how to specify the model in the first place. There is a need to be practice which accounts for the growing use of mathematical and statistical methods. However, as a systems becomes more complex, standard mathematical approaches may not be sufficient. In the field of ecology, the standard techniques involve discrete maps, and continuous models such as ODE's. The intent of this work is to present the mathematics necessary to study hybrids of these two models, then consider two case studies. In first case we con sider a coral reef with continuous change, except in the presence of hurricanes. The results of the data are compared quantitatively and qualitatively with simulation results. For the second case we consider a model for rabies with a periodic birth pulse. Here the analysis is qualitative as we demonstrate the existence of a strange attractor by looking at the intersections of the stable and unstable manifold for the saddle point generating the attractor. For both cases studies the introduction of a discrete event into a continuous system is done via a Dirac Distribution or Measure.

Show moreThe purpose of this research is to validate various forms of mathematical modeling of glioblastoma multiforme (GBM) expressed as differential equations, numerically. The first work was involved in the numerical solution of the reaction-convection model, efficacy of which is expressed in terms of survival time. It was calculated using simple numerical scheme for the standard-of-care treatment in clinics which includes surgery followed by the radiation and chemotherapy. Survival time using all treatment options increased significantly to 57 weeks compared to that of surgery close to 14 weeks. It was also observed that survival time increased significantly to 90 weeks if tumor is totally resected. In reaction-diffusion model using simple numerical scheme, tumor cell density patterns due to variation in patient specific tumor parameters such as net proliferation rate and diffusion coefficient were computed. Significant differences were observed in the patterns while using dominant diffusion and proliferation rate separately. Numerical solution of the tumor growth model under the anti-angiogenic therapy revealed some impacts in optimum tumor growth control however it was not significant.

Show moreTaurine is the second most abundant amino acid in the CNS after glutamate and its functions have been found largely related to intracellular calcium ([Ca2+]i) modulation, osmoregulation, membrane stabilization, reproduction and immunity. The action of taurine has also been implicated in neurotransmission and neuromodulation though its specific sites of action are not fully understood. Isolated retinal neurons from the larval tiger salamanders (Ambystoma tigrinum) were used as a model to study the neuromodulatory role of taurine in the CNS and to gain insights into its potential sites of action. A combination of techniques was used, including whole-cell patch clamp recording to study taurine's regulation of voltage-gated potassium (K+) and Ca2+ channels and Fluo-4AM Ca2+-imaging to study taurine's regulation of glutamate-induced [Ca2+] I,. Taurine was shown to suppress of glutamate-induced [Ca2+] l, in a dose dependent manner. This suppression was mostly sensitive to the glycine rece ptor antagonist Strychnine but insensitive to any GABA receptor antagonist. The remaining strychnine-insensitive effect was inhibited with the protein kinase A (PKA) inhibitor, PKI, suggesting that there was an additional metabotropic pathway. Moreover, using the protein kinase C (PKC) inhibitor, GF109203X, there was an enhancement in strychnine-insensitive taurine's regulation. Taurine inhibits voltage-gated Ca2+ channels in the retinal neurons and has a dual effect on voltage-gated K+ channels. Taurine causes an increase in K+ current amplitude which is further enhanced with PKI and blocked with GF109203X, suggesting that it is through a PKC-dependent pathway negatively controlled by PKA-dependent pathway., There is a suppression of K+ current by taurine with intracellular application of GF109203X, suggesting that the reduction is through a PKA-dependent pathway. With both PKC and PKA inhibitors there is no longer an enhancement in maximum amplitude but a shift of volt dependence on a hyperpolarizing direction. Taurine's enhancement of K+ current is blocked by the Kv1.3 subtype antagonist Margatoxin, with Kv1.3 accounting for the majority of delayed-rectifier sustained current in bipolar and amacrine cells, as well as 50% of ganglion cells. Interestingly, the enhancement of K+ current by taurine is blocked by 5HT2A antagonist MDL11939, suggesting that activation of PKC is through this metabotropic serotonin receptor subtype. The suppression of voltage-gated Ca2+ channels is reversed with a combination of MDL11939 and the 5HT1A antagonist NAN-190. These results provide the evidence that the natural effect of taurine in the retinal neurons might be dependent on the activation of both 5HT1A and 5HT2A receptors. The high apparent activity of taurine on 5HT receptors could have important implication for the actions of taurine in central brain in which taurine has been known to be beneficial for improving mental health, as well as learning and memory processes.

Show moreThe thesis objective is to develop protocols that provide analysis and interpretation for data from a pulsed laser serial scanning lidar system for underwater imaging. The specific focus is remotely observing marine organisms in the centimeter size range in the poorly understood mesopelagic realm of the ocean. The Unobtrusive Multi-Static Lidar Imager (UMSLI) system captures an expansive volume scan field with differentiating imaging resolution per planar slice, allowing precise assignment of location for organisms in the field of view. The multi-static highly collimated beams are recorded by a photo-multiplier tube receiver as a time lapse waveform of the returned energy flux, each waveform comprehensibly represents an image pixel in spatially and temporally. Complied lidar waveforms produce an array of returns which signify the magnitude of backscatter from varying sized particles across the observed volume. These volume scans are uniquely evaluated and transformed for each time bin through a processing method which extracts particle characteristics and statistics based on adaptive spatial and temporal techniques. The post processing method aims to greatly extend the capabilities of the lidar imaging system to extract particles. Results of the processing method are presented as particle counts and particle size distributions of the water columns during observed vertical migration periods. Methods are compared with other optical devices for validation, and results are interpreted to better understand the organism distribution in the mesopelagic and their behavior, with respect to diel vertical migrations.

Show moreHow do neuronal connectivity and the dynamics of distributed brain networks process information during bimanual coordination? Contemporary brain theories of cognitive function posit spatial, temporal and spatiotemporal network reorganization as mechanisms for neural information processing. In this dissertation, rhythmic bimanual coordination is studied as a window into neural information processing and subsequently an investigation of underlying network reorganization processes is performed. Spatiotemporal reorganization between effectors (limbs) is parameterized in a theoretical model via a continuously varying cross-talk parameter that represents neural connectivity. Thereby, effector dynamics during coordinated behavior is shown to be influenced by the cross-talk parameter and time delays involved in signal processing. In particular, stability regimes of coordination patterns as a function of cross-talk, movement frequency and the time delays are derived. On the methodological front , spatiotemporal reorganization of neural masses are used to simulate electroencephalographic data. A suitable choice of experimental control conditions is used to derive a paradigmatic framework called Mode Level Cognitive Subtraction (MLCS) which is demonstrated to facilitate the disambiguation between spatial and temporal components of the reorganization processes to a quantifiable degree of certainty. In the experimental section, MLCS is applied to electroencephalographic recordings during rhythmic bimanual task conditions and unimanual control conditions. Finally, a classification of reorganization processes is achieved for differing stability states of coordination: inphase (mirror) primarily entails temporal reorganization of sensorimotor networks localized during unimanual movement whereas spatiotemporal reorganization is involved during antiphase (parallel) coordination.

The study applied theories of the mind/body connection and stress frameworks for exploring the correlation between patients' hand temperatures and their reported levels of pain pre-operatively and post-operatively. Study findings indicated that there was significant within-subjects effect in hand temperature after biofeedback treatment across three points in time. But the study also found no difference between-subjects in hand temperature after biofeedback treatment across three points in time. Therefore, the sample proved to be heterogeneous. Further study was indicated with larger samples to demonstrate the analgesic effects of biofeedback in the management of acute pain.

At this scale the wavelength of the imaging field (usually near infra-red), is comparable to the size of the structures being imaged, which makes the use of ray optics invalid. A better understanding is needed to predict the result of introducing different media into the light path. We use Wolf's integral, which is capable of fulfilling these needs without the shortcomings of ray optics. We predict the effects of aberrating media introduced into the light path like glass cover-slips and then correct the aberration using the same method. We also create a method to predict aberrations when the interfaces of the media in the light-path are not aligned with the propagation direction of the wavefront.

Transgenic mouse models presenting the pathology observed in clinical patients with RCM have been generated previously and express the mutant cTnI in their hearts. RCM-linked mutations increase cardiac myofilament Ca2+ sensitivity and promote diastolic dysfunction in the heart. Previous studies using double transgenic mice (cTnI/R193H/ND) showed that ventricular relaxation is enhanced in the cTnI/R193H transgenic mice. In this study, another double transgenic mouse model, (cTnI/R193H/ND/KO), provides an avenue to investigate its rescuing effects on RCMlinked mutations in the cTnI /R193H/KO mouse. Use of molecular biological techniques, transgenic animal developments and murine echocardiography in this study has culminated into a greater understanding of RCM and diastolic dysfunction.

We reported that both therapeutic and prophylactic vaccines fail to protect and cure animals from disease due to negative effects of nicotine on DCs’ ability to generate effector T cells. We have been investigating whether vaccine formulated with TLR agonist(s) could potentially overcome the immunosuppressive effects of nicotine on human DC-NK cross-talk essential for effector T cell generation. Monocyte-derived DCs and nicDCs were stimulated with individual and combined TLR agonists prior to co-culture with purified T cells. The phenotypes and cytokine profiles of T cell were assessed using Flow Cytometry and ELISA, respectively. We found nicDCs cultured with TLR-8/7 alone or in combination with TLR-3 produce quantitatively and qualitatively similar IFN-γ producing effector T cells when compared to control DCs. Our data suggest that the addition of appropriate TLR agonist to vaccine formulation could potentially overcome the immunosuppression seen in smokers, thereby containing the spread of infectious disease to vulnerable population

Our laboratory is among the first to generate transgenic mouse models of RCM based on cardiac troponin I (cTnI) missense mutations. In this study, transgenic mice that suffer from RCM have been generated to understand the factors behind the diastolic dysfunction associated with that myocardial disease.... The information obtained from this study allows a better understanding of the role of troponin in RCM and the factors behind the physiopathology of the disease. It will also offer a therapeutic strategy taking into account the physiological characteristic of RCM.

The primary purpose of this study was to investigate the effect of acute high-intensity interval exercise (HIIE) vs. continuous moderate-intensity exercise (CME) on serum CTRP9 and brachial FMD responses in obese and normal-weight subjects. Sixteen participants (9 obese and 7 normal-weight) completed HIIE and CME in a randomized fashion. Show moreThe primary purpose of this study was to investigate the effect of acute high-intensity interval exercise (HIIE) vs. continuous moderate-intensity exercise (CME) on serum CTRP9 and brachial FMD responses in obese and normal-weight subjects. Sixteen participants (9 obese and 7 normal-weight) completed HIIE and CME in a randomized fashion. Our results showed a significant time effect for CTRP9 immediately following acute HIIE and CME in both groups. Furthermore, both significant treatment by time and group by time interactions for FMD were observed following both exercise protocols, with greater CME-induced FMD response in obese subjects than normal-weight subjects. Additionally, a positive correlation in percent change (baseline to peak) between CTRP9 and FMD was observed following acute CME. These findings support acute CME for improvement of endothelial function in obesity. Furthermore, the novel results from this study provide a foundation for additional examination of the mechanisms of exercise-mediated CTRP9 on endothelial function.

Show moreIn the past 25 years, Palm Beach County has acquired more than 31 million acres of public preserves consisting of numerous native habitats that are home to threatened and endangered flora and fauna. Both endemic habitats and plant species benefit from expansion and connectivity through the adoption of native plants in surrounding areas. The next step in conserving these protected, larger areas is to identify ways to connect them in order to reduce their isolation and improve their likelihood of continued ecosystem health. This study aims to perform an analysis using a GIS database of Palm Beach County's Eastern urban areas to determine additional vegetation that currently exists on public lands and private preserves and to analyze and classify the vegetation for its potential conservation role, either as ; preserves, buffers, corridors or clusters connecting and enhancing existing natural areas.
